摘要
目的:考察太子参均一多糖对大鼠小肠α-糖苷酶活性影响,探讨其降血糖途径。方法:由大鼠分离小肠α-糖苷酶,以对硝基酚-α-葡萄糖苷(pNPG)为底物,利用α-葡萄糖苷酶将其分解成pNP及葡萄糖,而pNP在碱性条件下显色的原理测定酶活力,以考察太子参均一多糖对酶活性的影响。结果:太子参均一多糖HP_(0.5MSC-F)和HP_(H-1-2),浓度为400μmol/L时对大鼠小肠α-糖苷酶活性抑制率分别为19.00%和28.07%。结论:两种太子参均一多糖对α-葡萄糖苷酶活性具有一定的抑制作用。
Objective To investigate the effect of homogenous polysaccharide of Pseudostellaria heterophylla on the activity of α-glucosidase in the small intestine of rats and to explore its hypoglycemic pathway. Methods The α-glucosidase was isolated from rat small intestine. The α-glucosidase was decomposed into pNP and glucose by α-glucosidase using p-nitrophenol α-glucoside (pNPG) as substrate, and the activity of pNP was determined by the principle of color development under alkaline conditions. The effect of homogeneous polysaccharide of Pseudostellaria heterophylla on enzyme activity was investigated. Results The α-glucosidase activity inhibitory rates of homopolysaccharide of HP 0.5m and HP H-1-2 were 19.00% and 28.07% respectively at the concentration of 400 μmol/L. Conclusion Two homogenous polysaccharides of Pseudostellaria heterophylla have a certain inhibitory effect on α-glucosidase.
