金属离子对茯苓液态发酵合成三萜的调控效应研究 被引量:1

摘要 本文从金属离子种类、添加浓度、添加时间角度对金属离子调控液态深层发酵体系茯苓三萜合成的效应进行了系统分析,并初步探索了基于金属离子调控了茯苓菌液态发酵合成茯苓三萜的工艺放大。结果显示,在发酵48 h添加200μM锌离子可使茯苓三萜产量达27.22 mg/L,为对照组的2.34倍;10 L罐放大工艺试验显示在发酵76 h茯苓三萜产量即可达到25.19 mg/L,发酵大幅缩短。这些数据为强化液态发酵体系茯苓三萜生物合成的相关研究提供了必要参考。 For improving triterpenoids production in Poria cocos submerged fermentation system, effects of differentiation metal ions, ion concentration and ion addition time on triterpenoids production were explored. Besides, primary scale-up of Poria cocos fermentation process was also studied in a 10-L stirred bioreactor based on ion regulation strategy. Results showed that a maximum triterpenoids production in flask level can reach 27.2 mg/L when 200 μM zinc sulfate were added to the broth after 48 h fermentation. This indicated that a 2.34 fold triterpenoids production process was realized under zinc ion regulation. In 10-L bioreactor, the triterpenoids concentration can reach 25.19 mg/L only after 76 h after fermentation beginning, implying a big shortening of fermentation cycle. This reach can provide an important reference for improving biosynthesis efficiency of Poria cocos in submerged fermentation system.
