摘要
本文采用胰蛋白酶-木瓜蛋白酶连续双酶解法获得甲鱼肉蛋白酶解物(Pelodiscus sinensis protein hydrolysate, PsPH)及其超滤分级组分(大于10 kDa、3~10 kDa和小于3 kDa),并以DPPH自由基、ABTS自由基和羟基自由基清除能力以及氧自由基吸收能力为评价指标分析了其抗氧化活性进行。结果表明,甲鱼肉蛋白酶解物的三个超滤分级组分均表现出较好的ABTS自由基清除能力和氧自由基吸收能力。分子量小于3 kDa的组分还显示出较好的羟基自由基清除能力,在浓度为4mg/mL时对羟基自由基的清除率大于85%。这些结果说明采用胰蛋白酶-木瓜蛋白酶连续双酶解法制备的甲鱼肉蛋白酶解物具有较好的抗氧化活性。
The protein hydrolysate of Chinese soft-shelled turtle(Pelodiscus sinensis) meat(PsPH) was prepared by double enzymatic digestion sequentially with trypsin and papain, followed by fractionation with ultrafiltration, which yielded >10 kDa, 3~10 kDa and <3 kDa fractions. The scavenging ability of the hydrolysate and its ultrafiltration fractions against DPPH, ABTS and ·OH free radicals as well as their absorption capacity of oxygen free radicals were used to evaluate their antioxidant activity. All three fractions of the hydrolysate showed ABTS scavenging effect and oxygen radical absorption capcity. The <3 kDa fraction also showed strong scavenging ability against ·OH free radicals, which was >85 % at 4 mg/ml. Together, these data demonstrate that the hydrolysate obtained by trypsin-papain double hydrolysis from Chinese soft-shelled turtle has good antioxidant activity.
