血管抑素对大鼠角膜碱烧伤后瘢痕形成的作用研究

摘要 目的:研究血管抑素(AS)对大鼠角膜碱烧伤后瘢痕形成的作用。方法:取100只SD大鼠,复制角膜碱烧伤模型,随机分为4组:AS1组、AS2组、地塞米松组和生理盐水组,每组25只,分别用10μg/mL AS、20μg/mL AS、1mg/mL地塞米松和生理盐水点眼,4次/d。于碱烧伤后1d、3d、7d、14d、21d通过裂隙灯观察角膜混浊度,苏木精—伊红(HE)染色及光镜下观察炎症细胞数目,电镜下观察角膜超微结构变化。结果:碱烧伤后3~21d,AS1组、AS2组及地塞米松组角膜混浊度评分均优于生理盐水组,且AS2组评分优于地塞米松组,差异有统计学意义(P<0.05);碱烧伤后1~21d,AS1组、AS2组及地塞米松组炎症细胞数均少于生理盐水组,且AS2组炎症细胞数少于地塞米松组,差异均有统计学意义(均P<0.05)。结论:AS能有效抑制大鼠角膜碱烧伤后瘢痕形成,且效果优于地塞米松。 Objective: To study the effect of angiostatin (AS) on scar formation after corneal alkali burn in rats. Methods: A total of 100 Sprague-Dawley rats were divided into four groups and corneal alkali burn models were established (n=25 per group): AS1 group (10 μg/mL AS),AS2 group (20 μg/mL),dexamethasone (1 mg/mL) group and normal saline group.Rats were treated with different doses of AS,dexamethasone or normal saline eye drops four times daily.On day 1,3,7,14,21 after alkali burn,the corneal opacity was observed by slit lamps,and inflammatory cells number was counted under light microscopy.Corneal ultrastructure was observed by electron microscopy. Results: The corneal opacity score of AS1,AS2 and dexamethasone groups was better than that of normal saline group on day 3-21 after alkali burn,and the score of AS2 group was better than that of dexamethasone group (P<0.05).The number of inflammatory cells in AS1,AS2 and dexamethasone groups was less than that of normal saline group after 1-21 days of alkali burn,and the inflammatory cells number in AS2 group was less than that in dexamethasone group (P<0.05). Conclusion: AS could effectively restrain the formation of corneal scar after alkali burns in rats,and the effect was superior to dexamethasone.
