结直肠癌患者遗传易感性与外周血双向调节蛋白基因多态性的关系

Relationship between gene polymorphism of amphiregulin in peripheral blood and genetic susceptibility of patients with colorectal cancer in Shanghai

目的:探讨结直肠癌(CRC)患者外周血中双向调节蛋白(AREG)基因多态性与CRC遗传易感性的关系。方法:选取2013年7月至2017年6月上海中医药大学附属曙光医院收治的240例CRC患者为观察组,同期选取240例健康志愿者为对照组,对比两组血清AREG水平及外周血中AREG基因rs161511、rs2291715位点基因型分布和等位基因频率,Logistic回归分析法分析rs161511、rs2291715多态性与CRC遗传易感性关系。结果:观察组血清AREG水平显著高于对照组(P<0.05);两组受试者AREG基因rs161511位点基因型等级分布比较无明显差异(P>0.05);两组受试者rs2291715位点基因型等级分布比较,差异有统计学意义(P<0.05),且rs2291715位点C等位基因频率显著低于对照组(P<0.05);rs2291715位点CC、CG、GG基因型及C、G等位基因可能是CRC遗传易感性影响因素(P<0.05)。结论:CRC患者血清AREG异常高表达,CRC患者遗传易感性与AREG基因rs2291715位点多态性有关,与rs161511位点多态性可能无关。

Objective: To investigate the relationship between gene polymorphism of amphiregulin (AREG) in peripheral blood and genetic susceptibility of patients with colorectal cancer (CRC) in Shanghai. Methods: A total of 240 CRC patients were selected as observation group from July 2013 to June 2017 in our hospital.240 healthy subjects were selected as control group during the same period.The level of serum AREG,the frequencies of gene-types distribution and alleles in rs161511 and rs2291715 sites in peripheral blood were compared.The relationship between rs161511,rs2291715 polymorphism and CRC were analyzed by logistic regression analysis method. Results: The level of serum AREG of the observation group was significantly higher than that of the control group (P<0.05).There was no significant difference in the distribution of the alleles in rs161511 site between the two groups (P >0.05).There was significant difference in the distribution of the alleles in rs2291715 site between the two groups (P<0.05),and the frequency of C allele in rs2291715 site of the observation group was significantly lower than that of the control group (P<0.05).The CC,GG,GC gene-types and the C,G alleles in rs2291715 site may be the influencing factors of genetic susceptibility of CRC (P<0.05). Conclusion: The serum AREG level of CRC patients was highly expressed.The rs2291715 site polymorphism may participate in genetic susceptibility of patients with CRC in Shanghai,but it may not be related to the rs161511 site polymorphism.
