摘要
目的观察针刺对实验性自身免疫性脑脊髓炎(EAE)小鼠脊髓钙离子接头蛋白-1(IBA-1)、环氧合酶-2(COX-2)及血清肿瘤坏死因子-α(TNF-α)水平的影响,探讨其可能的作用机制。方法将36只C57BL/6小鼠随机分为4组,正常组小鼠不予处理,其余小鼠均制作EAE模型。造模后第14日开始,正常组和模型组小鼠每日抓取1次,激素组小鼠给予醋酸泼尼松灌胃,针刺组小鼠针刺双侧"肾俞""足三里""大椎"。造模后28 d取材,检测小鼠血清TNF-α含量及脊髓COX-2、IBA-1阳性表达。结果与正常组比较,模型组小鼠神经功能评分及TNF-α、COX-2、IBA-1水平均明显升高(P<0.01);与模型组比较,针刺组和激素组小鼠神经功能评分和TNF-α、COX-2、IBA-1水平均明显降低(P<0.01);与针刺组比较,激素组小鼠神经功能评分和COX-2、IBA-1水平均明显降低(P<0.05,P<0.01)。结论针刺可降低EAE小鼠炎症反应,减轻脊髓神经损伤,其机制可能与降低血清TNF-α含量,减少脊髓COX-2、IBA-1阳性表达有关。
Objective To observe the effects of acupuncture on the levels of IBA-1,COX-2 in spinal cord and serum TNF-αof experimental autoimmune encephalomyelitis(EAE)mice model;To clarify the mechanism of acupuncture treatment.Methods Thirty-six C57BL/6 mice were randomly divided into 4 groups.The normal group of mice were not immunologically molded,and the others were injected to replicate the EAE model.Since the 14th day after modeling,the normal group and the model group mice were taken once a day.Hormone group received prednisone acetate for gavage.Acupuncture group received acupuncture at"Shenshu","Zusanli",and"Dazhui".At the 28th day after of modeling,the content of serum TNF-αand the positive expressions of COX-2 and IBA-1 in the spinal cord were detected.Results Compared with the normal group,neurological function score,TNF-α,COX-2 and IBA-1 in the model group increased(P<0.01).Compared with the model group,the neurological function scores and TNF-α,COX-2 and IBA-1 levels in the acupuncture group and the hormone group were significantly lower(P<0.01).Compared with the acupuncture group,the neurological function score and COX-2 and IBA-1 levels in the hormone group were significantly lower(P<0.05,P<0.01).Conclusion Acupuncture can reduce the inflammatory response of EAE mice and alleviate spinal nerve injury.The mechanism may be related to lowering serum TNF-αcontent and reducing the positive expressions of COX-2 and IBA-1 in spinal cord.
