摘要 目的探讨急性湿疹大鼠皮肤屏障功能的变化及机制。方法选择SD大鼠20只,随机分为正常组、模型组各10只。除正常组外,模型组采用二硝基氯苯(DNCB)在右背部皮肤建立急性湿疹模型。造模成功后,取大鼠右背部皮肤,采用HE染色法观察炎细胞浸润情况;采用皮肤表皮水分丧失测定仪测定经皮水分丢失量(TEWL);采用免疫组化法观察蛋白酶激活受体2(PAR-2)蛋白表达;采用RT-PCR法测定皮肤人类中间丝聚合蛋白(FLG) mRNA表达。结果模型组大鼠出现明显急性湿疹症状,皮肤TEWL、PAR-2含量增加,FLG mRNA表达降低(P均<0. 01)。结论急性湿疹的发生可能是通过降低FLG基因表达,增加PAR-2含量,增加TEWL,从而破坏皮肤屏障功能、诱发急性湿疹。 Objective To investigate the changes of skin barrier function of rats with acute eczema and its possible mechanism. Methods Twenty SD rats were randomly divided into the normal group and model group. In addition to the normal group, we used dinitrochlorobenzene (DNCB) to establish the acute eczema models on the right dorsal skin of rats in the model group. After the success of modeling, the right back skin of rats was taken, and the infiltration of inflammatory cells was observed by HE staining. The transepidermal water loss (TEWL) was measured by skin epidermal water loss tester. The protein expression of proteinase activated receptor 2 (PAR-2) was observed by immunohistochemistry. The expression of filaggrin (FLG) mRNA was determined by RT-PCR. Results In the model group, the symptoms of acute eczema appeared, the content of TEWL and PAR-2 increased, and the expression of FLG mRNA decreased (all P <0.01). Conclusion Acute eczema may occur by reducing FLG gene expression, increasing PAR-2 and TEWL, thereby destroying skin barrier function and inducing acute eczema.
