摘要
目的探讨钙调蛋白对通道的调节功能,分析钙调蛋白与Na_V1.1 IQ在不同Ca^(2+)离子浓度下的结合情况,进一步探讨钙调蛋白和Na_V1.1结合的Ca^(2+)依赖性。方法应用pull-down技术检测不同Ca^(2+)离子浓度下Na_V1.1 IQ与钙调蛋白及其突变体的结合情况。结果 CaM野生型与Nav1.1 IQ基序的结合随着钙浓度和CaM浓度的增加而增高,而CaM 1234与Nav1.1 IQ的结合也随着CaM_(1234)浓度的增加而增高,但在不同钙浓度下的结合无变化。结论 CaM野生型和CaM_(1234)对电压门控性钠通道Nav1.1具有调节作用。
Objective To study the modulation of CaM on Nav1.1, detecting the binding of CaM on Nav1.1 IQ at different calcium concentrations to further explore the Ca^2+-dependent binding of CaM on Nav1.1. Methods Pull-down assay was used to detect the binding of CaM1234 on Nav1.1 under different calcium concentrations. Results The binding ability of CaM1234 on Nav1.1 was increased with increasing concentrations of Ca^2+/CaM. As the binding ability of CaM1234 on Nav1.1 increased with increasing concentrations of CaM1234, but the binding ability maintains at the same level at different calcium concentrations. Conclusion Both wild-type CaM and CaM1234 have regulatory effect on Nav1.1.
