六苓扶正方对环磷酰胺致免疫低下小鼠的免疫增强作用 被引量:7

摘要 目的:研究六苓扶正方对环磷酰胺致免疫低下小鼠免疫功能的影响。方法:将72只Babl/c小鼠随机分为6组:正常对照组,模型对照组,六苓扶正方低、中、高剂量组(1,2,4 g·kg^(-1))和阳性对照组(人参蜂王浆,2 g·kg^(-1)),每组12只。六苓扶正方各剂量组及阳性对照组分别灌胃给予相应药物,正常对照组和模型对照组给予等量蒸馏水,1次/d,连续30 d。给药第25d和26d,以40 mg·kg^(-1)腹腔注射环磷酰胺复制免疫低下模型,观察小鼠体质量及精神状态变化,考察六苓扶正方对血常规、免疫球蛋白G(IgG)、细胞白介素-2(IL-2)、胸腺指数、脾脏指数、脾淋巴细胞增殖及自然杀伤(NK)细胞活性的影响。结果:各组小鼠体质量差异无统计学意义(P>0.05),正常对照组小鼠精神及饮食状态良好,模型对照组小鼠精神萎靡,但饮食无明显变化,给予六苓扶正方干预后小鼠精神状态有明显改善。与正常对照组相比,模型对照组的白细胞计数(WBC)、红细胞计数(RBC)、血红蛋白浓度(Hb)、血小板计数(Plt)、IgG和IL-2水平、脾脏指数和胸腺指数、脾淋巴细胞增殖、NK细胞活性显著降低(P<0.05或P<0.01)。与模型对照组相比,六苓扶正方中、高剂量组的WBC和IgG水平显著升高(P<0.05或P<0.01),六苓扶正方高剂量组脾淋巴细胞增殖和NK细胞活性显著升高(P<0.05),六苓扶正方作用均呈剂量依赖性。六苓扶正方低剂量组的WBC和IgG水平及六苓扶正方高剂量组的脾淋巴细胞增殖和NK细胞活性与阳性对照相比差异有统计学意义(P<0.05或P<0.01)。结论:六苓扶正方具有增强免疫低下小鼠免疫功能的作用。 Objective: To explore the effect of Liulingfuzheng recipe( LLFZ) on the mice with immunosuppression induced by cyclophosphamide. Methods: Totally 72 Babl/c mice were randomly divided into 6 groups: the normal control group,model control group,low,medium and high dose groups of Liuling Fuzheng recipe( 1,2 and 4 g·kg^-1) and the positive control group( ginseng Royal jelly,2 g·kg-1),with 12 mice in each group. Liuling Fuzheng recipe groups and the positive control group were given corresponding drug by gavage,and the normal control group and the model control group were given the same amount of distilled water,once a day for 30 consecutive days. On the 25 th and 26 th days of administration,40 mg·kg^-1 cyclophosphamide was injected intraperitoneally to replicate the immunodeficiency model. The changes of body weight and mental status of mice were observed,and the effects of Liuling Fuzheng recipe on blood routine,immunoglobulin G( Ig G),interleukin-2( IL-2),thymus index,spleen index,spleen lymphocyte proliferation and natural killer( NK) cell activity were investigated. Results: There was no significant difference in body mass among the groups( P > 0. 05). The mice in the normal control group had good mental and dietary status,the mice in the model control group were depressed,but their diet did not change significantly,and the mental state of the mice was improved significantly after the intervention of Liuling Fuzheng recipe. Compared with the normal control group,the WBC,RBC,Hb,Plt,IgG and IL-2 levels,spleen index and thymus index,spleen lymphocyte proliferation and NK cell activity in the model control group were significantly decreased( P < 0. 05 or P < 0. 01). Compared with the model control group,the levels of WBC and IgG in the medium and high dose groups of Liuling Fuzheng recipe were significantly higher( P < 0. 05 or P < 0. 01),the proliferation of splenic lymphocyte and NK cell activity in the high dose group of Liuling Fuzheng recipe were significantly higher( P < 0. 05),and the effects of Liuling Fuzheng recipe were dose-dependent. The levels of WBC and IgG in the low dose group of Liuling Fuzheng recipe and the proliferation of splenic lymphocyte and NK cell activity in the high dose group of Liuling Fuzheng recipe were significantly different from those in the positive control group( P < 0. 05 or P < 0. 01). Conclusion: LIFZ can enhance the immune function of immunocompromised mice.
