摘要
采用闪式提取法提取杨树花总黄酮,并通过单因素试验和响应面优化试验,获得最佳提取工艺。同时,采用实时荧光定量RT-PCR方法检测杨树花提取物对蓖麻油诱导腹泻小鼠肠黏膜各种促炎因子相对表达量的影响,对杨树花提取物抗蓖麻油诱导小鼠腹泻作用进行了研究。结果显示,杨树花总黄酮的最佳提取工艺条件为:提取电压为113 V,乙醇体积分数为53.8%,提取时间为85 s,液料比为42.2∶1。在此条件下,总黄酮得率达到54.21 mg/g,与模型预测值54.59 mg/g相近。结果表明,此方法稳定可靠,提取率高,适用于杨树花总黄酮的提取。杨树花提取物对蓖麻油诱导腹泻小鼠具有较好的抗腹泻作用,可以明显降低小鼠肠黏膜促炎因子TNF-α、IL-6以及IL-1β的相对表达量,表明杨树花抗腹泻作用可能与降低促炎因子有关,为杨树花的综合开发利用提供了理论依据。
The total flavonoids of Flos Populi were extracted by flash extraction method,and the best extraction process was obtained by single factor experiment and response surface optimization experiment.At the same time,real-time quantitative RT-PCR was used to detect the effect of Flos Populi extract on the relative expression of pro inflammatory cytokines in the int estinal mucosa of castor oilinduced diarrhea in mice,to study its anti-diarrhea effect of Flos Populi extract.The results showed that the optimum extraction conditions for total flavonoids in Flos Populi were:extract ion voltage of 113 V,ethanol concentration of 53.8%,extraction time of 85 s,and liquid-to-material ratio of 42.2:1.Under these conditions,the total flavonoid yield reached 54.21 mg/g,which was similar to the model predicted value of 54.59 mg/g.The method was stable and reliable,and had high extraction rate,and was suitable for the extraction of total flavonoids from Flos Populi.The extract of Flos Populi had good anti-diarrhea effect on the castor oil-induced diarrhea in mice,and had sinignifiant effect on the decrease of relative expression of proinflammatory factors TNF-α,IL-6 and IL-1β in the intestinal mucosa of mice,indicating that the anti-diarrhea effect of Flos Populi extract may be related to the reduction of proinflammatory factors.This study provided theoretical foun dation for the comprehensive development and utilization of Flos Populi.
