我国3个主要民族HIV感染者基因组中人内源性逆转录病毒K113和K115基因分布研究 被引量:2

Different distributions of HERV-K113 and K115 genes found in HIV-1 infected individuals of different ethnics in China

摘要 目的分析人内源性逆转录病毒(HERV) K113和K115基因在我国不同民族HIV感染者基因组中的分布,为进一步研究其与HIV感染和复制的关系奠定基础。方法收集我国广西、云南、四川和河南HIV感染者的血液样本,提取基因组DNA,通过PCR扩增及核酸电泳检测,判断样本基因组中HERV-K113、HERV-K115基因插入状况。采用2检验和Fisher精确检验分析HERV-K113、HERV-K115基因在不同民族、地区HIV感染者中的分布是否存在差异。结果收集我国汉、彝、壮族3个民族共504份HIV感染者血液样本(汉族238例,彝族175例,壮族91例),基因组中HERV-K113和HERV-K115插入率分别为30. 0%和4. 8%;不同民族HERV-K113插入率分别为39. 6%(壮族)、30. 3%(汉族)和24. 6%(彝族); HERV-K115插入率分别为7. 7%(壮族)、5. 0%(汉族)和2. 9%(彝族)。HERV-K113在不同民族的分布存在显著性差异(P <0. 05)。结论首次阐明HERV-K113、HERV-K115基因在我国不同民族HIV感染者基因组中的频率分布,为进一步研究其与HIV感染及疾病进程的相关性奠定了基础。 Objective To demonstrate the genetic distributions of human endogenous retrovirus(HERV)K113 and K115 among HIV infected individuals from different areas or ethnics in China.Methods Peripherial blood was sampled and genomic DNA was extracted from HIV-1 positive individuals in China.The presence or absence of HERV-K113 and HERV-K115 was determined with PCR primers spanning the preintegration sites and sites within the viral sequences.Results A total of 504 participants were enrolled into the study.Among them,238 were from the Han Chinese,175 from Yi ethnic minority,and 91 from Zhuang ethnic minority.The insertion frequencies of HERV-K113 and HERV-K115 were30.0%and 4.8%,respectively.The distributions of HERV-K113 were significantly different between ethnics and regions.The highest frequency of K113 was identified in Zhuang(39.6%),followed by Han(30.3%)and Yi(24.6%)within HIV-1 positive individuals in China.HERV-K115 was found in 7.7%of Zhuang beople,5.0%of Han and 2.9%of Yi.Conclusion To date,there has been no report on HERV-K113 and HERV-K115 prevalence in HIV-1 infected individuals in China.This study will provide information for detecting the relationships between presence of the HERVs and disease procession of HIV-1 infected individuals.
