一测多评法测定甜荞中6种酚类成分 被引量:2

Simultaneous quantitative analysis of six componentsin by quantitative analysis of multi-components by single marker method in buckwheat

摘要 建立了一测多评法(QAMS)测定甜荞中6种酚类成分的含量。采用超高效液相色谱法,色谱柱为ACQUITYUPLC~?BEHC18(1.7μm,2.1 mm×50 mm),甲醇-0.2%冰乙酸水溶液为流动相梯度洗脱,检测波长254 nm,流速0.2 mL/min,柱温35℃;以芦丁为内参物,建立其与儿茶素、表儿茶素、阿魏酸、槲皮苷、槲皮素的相对校正因子,采用一测多评法和外标法(ESM)同时计算各待测成分的含量,并进行比较。结果表明,10种甜荞品种中儿茶素、表儿茶素、阿魏酸、芦丁、槲皮苷、槲皮素的含量可以用一测多评法进行测定,一测多评法测定的结果与外标法无明显差异(P>0.05),并且10个品种中红花甜荞的6种酚类成分量最高,为112.94μg/g;最低是丰甜1号,含量为54.60μg/g。文章建立的一测多评法操作简单、重复性好,可为甜荞的质量评价提供参考。 A quantitative method of analysis of multi-components by single marker(QAMS) method for the quantification of six polyphenols in buckwheat was established. Samples were analyzed by Ultra Performance Liquid Chromatography(UPLC) system, equipped with a ACQUITYUPLC?BEHC18(1.7 μm,2.1 mm×50 mm) chromatographic column, with the mobile phase of methanol-0.2% glacial acetic acid solution at a flow rate of 0.2 mL/min. The column temperature and the detection wavelength set at 35 ℃and 254 nm respectively. The rutin was used as the internal reference to determine the relative correction factors(RCFs) of other five components, and their contents were calculated by their RCFs at the same time, then compared with the external standard method(ESM). No significant differences were found in the quantitative results of six polyphenols in 10 varieties of buckwheat determined by ESM and QAMS(RSD<5%). The highest total content of 6 phenolic substances in 10 varieties of buckwheat is Honghua(112.94 μg/g), The lowest is Fengtian(54.60 μg/g). It is feasible and suitable to evaluate the quality of buckwheat by QAMS.
