炭疽病是大薯种植过程中所遭受的最严重病害。为了探究PR1基因家族对炭疽菌的响应情况,本研究在前期转录组的基础上筛选了6个PR1基因,设置5组处理;炭疽菌处理、MeJA处理、SA处理、MeJA+炭疽菌处理、SA+炭疽菌处理,分别在处理1 d、2 d、3 d、4 d、5 d采用荧光定量PCR进行6个基因表达量差异分析。结果显示6个不同PR1基因在炭疽菌处理下都会使得其表达量增高,炭疽菌诱导下PR1-1基因5 d时表达量为6个基因中最高,达到峰值。炭疽菌+MeJA诱导下PR1-6表达量较高,4 d时达到峰值。炭疽菌+SA诱导下PR1-9在4 d表达量达到峰值。Me JA诱下PR1-6基因表达量较高,2 d时达到峰值。SA诱导下PR1-1基因2 d时表达量达到峰值,于6个基因中最高。由此可见,不同PR1基因对炭疽菌以及不同激素处理响应不同。
Anthracnose is the most serious disease in Dioscorea alata. In order to research the response of PR1 gene family to colletotrichum asianum, MeJA and SA, six PR1 genes were selected on the basis of early transcription. This research had five treatments including, colletotrichum asianum treatment, Me JA treatment, SA treatment, MeJA with colletotrichum asianum treatment and SA with colletotrichum asianum treatment. Fluorescence quantitative reaction was used to analyze the differences of six genes expression at 1 d, 2 d, 3 d, 4 d and 5 d after5 treatments. The results showed that the expression of six different PR1 genes increased under the treatment of colletotrihum asianum. PR1-1 was the highest expression of PR1 gene and reached its peak at 5 d after colletotrichum asianum treatment. The expression of PR1-6 was higher than other PR1 genes under MeJA with colletotrichum asianum treatment and reached its peak at 4 d. The expression of PR1-9 reach its peak at 4 d after SA with colletotrichum asianum treatment. The expression of PR1-6 gene induced by MeJA was high and reached its peak at 2 d. The expression of PR1-1 gene reached its peak at 2 d after SA treatment, which was the highest among the six genes, at same time. Therefore, different PR1 genes responded differently to different hormones and colletotrichum asianum treatments.
