血府逐瘀汤对冠心病动物模型血脂和心肌酶学的影响 被引量:11

Effects of Xuefuzhuyu Decoction on Blood Fat and Myocardial Enzymes in the Coronary Heart Disease Rat Model

摘要 目的观察血府逐瘀汤对冠心病动物模型血脂和心肌酶学的影响。方法 SPF级SD大鼠随机分为对照组、模型组、假手术组、血府逐瘀汤低剂量、中剂量、高剂量6组,每组10只。模型组、低剂量、中剂量、高剂量组采用左冠状动脉放置缩窄环方法,制备冠心病模型。低剂量、中剂量、高剂量组以血府逐瘀汤按10、20、40 g/kg灌胃,1次/d,连续21 d。对照组、模型组、假手术组则给予等体积生理盐水。检测血管紧张素Ⅱ(AngⅡ)、活性氧(ROS)、三酰甘油(TG)、血清总胆固醇(TC)、低密度脂蛋白胆固醇(LDL-C)、高密度脂蛋白胆固醇(HDL-C)、天门冬氨酸氨基转移酶(AST)、丙氨酸转氨酶(ALT)、肌酸激酶(CK)、乳酸脱氢酶(LDH)水平及NOX2蛋白表达;HE染色观察心肌细胞形态学变化。结果与对照组比较,模型组AngⅡ、ROS、TG、TC、LDL-C、AST、ALT、CK、LDH水平及NOX2表达升高或显著升高(P<0.05,P<0.01),而HDL-C水平显著下降(P<0.01);与模型组比较,中剂量、高剂量组AngⅡ、ROS、TG、TC、LDL-C、AST、ALT、CK、LDH水平及NOX2表达降低或显著降低(P<0.05,P<0.01),而HDL-C水平显著升高(P<0.01)。结论血府逐瘀汤具有减轻氧化应激损害的效果,同时也可降低血脂水平,部分体现其"活血化瘀"的作用。 Objective To investigate the effects of Xuefuzhuyu decoction on blood fat and myocardial enzymes in the rats with coronary heart disease. Methods SPF grade SD rats were randomly divided into control group, model group, sham group, and Xuefuzhuyu decoction by low-dose, medium-dose and high-dose groups. Left coronary artery implanted constrictor was used to establish the model of coronary heart disease in the model group, the low-dose, the medium-dose and the high-dose groups. The rats in the low-dose, the medium-dose and the high-dose groups received intragastric administration of Xuefuzhuyu decoction of 10, 20 and 40 g·kg-1 once a day for continuous 21 days, respectively, while those in the control group, the model group and the sham group received same volume normal saline. The levels of Ang II, ROS, TG, TC, LDL-C, HDL-C, AST, ALT, CK and LDH, and the expression of NOX2 were measured. The cardiac muscle morphological features were observed through HE stained. Results Compared with the control group, in the model group the levels of AngII, ROS, TG, TC, LDL-C, AST, ALT, CK and LDH, and the expression of NOX2 protein were significantly increased(P<0.05), while the level of HDL-C was significantly reduced(P<0.01). Compared with the model group, in the medium-dose and the high-dose groups the levels of AngII, ROS, TG, TC, LDL-C, AST, ALT, CK and LDH, and the expression of NOX2 protein were significantly reduced(P<0.05), while the level of HDL-C was significantly increased(P<0.01). Conclusion Xuefuzhuyu decoction could alleviate oxidative stress injury and reduce the levels of blood fat, which partly embodies its effect on promoting blood circulation and removing blood stasis.
