葛根黄酮对癫痫大鼠海马NF-κB mRNA和IL-10mRNA表达的影响

Effects of Pueraria Flavones on Expressions of Neclear Factor-κB mRNA and IL-10 mRNA in Hippocampus of Epileptic Rats

摘要 目的:探讨葛根黄酮对癫痫大鼠脑神经的保护作用。方法:制备葛根黄酮溶液,通过腹腔注射亚惊厥剂量戊四氮构建癫痫大鼠模型,将大鼠分为对照组、PZT模型组和葛根黄酮干预组,在7、14、21、28 d分别取大鼠海马组织进行实时荧光定量PCR检测NF-κB mRNA和IL-10mRNA的表达。结果:随着给药时间的延长,与对照组比较,PZT模型组各时间点NF-κB mRNA和IL-10 mRNA的表达增高,差异有统计学意义(P<0.01),葛根黄酮干预组在21、28 d NF-κB mRNA和IL-10 mRNA的表达量增高,差异有统计学意义(P<0.01);且与模型组比较,干预组在21、28 d NF-κB mRNA和IL-10 mRNA的表达量降低,差异有统计学意义(P<0.01)。结论:葛根黄酮溶液对癫痫大鼠脑损伤具有一定的保护作用。 Objective:To investigate the protective effects of Pueraria Flavones on brain nerve in epileptic rats.Method:Pueraria Flavones solution was prepared,and the epileptic rat model was constructed by intraperitoneal injection of Pentylenetetrazol.The rats were divided into control group,PZT model group and Pueraria Flavone intervention group.The hippocampal tissues of rats were extracted on 7 d,14 d,21 d and 28 d respectively for real-time quantitative PCR detection of the expression of NF-κB mRNA and IL-10 mRNA.Result:With the extension of drug administration time,compared with the control group,the expression of NF-κB mRNA and IL-10 mRNA in the PZT model group increased at each time point,the differences were statistically significant(P<0.01).And compared with the model group,the expression level of NF-κB mRNA and IL-10 mRNA in the Pueraria Flavones intervention group on 21 d and 28 d were decreased,the differences were statistically significant(P<0.01).Conclusion:Pueraria Flavones Solution has a protective effect on brain injury of epileptic rats.
