高效液相色谱法同时测定加工肉制品中防腐剂和抗氧化剂 被引量:2

Simultaneous Determination of 11 Preservatives and Antioxidants in Processed MeatUsing High-performance Liquid Chromatography with Diode Array Detector

摘要 针对加工肉制品基质复杂,采用固相萃取的样品前处理方式,建立二极管阵列高效液相色谱法梯度洗脱同时测定加工肉制品中的防腐剂和抗氧化剂。采用Xtimate C18(4.6 mm×250 mm,5μm)色谱柱分离,以甲醇-0.02 mol/L乙酸铵溶液为流动相梯度洗脱,标准曲线法定量。防腐剂、抗氧化剂的线性范围为0-10.0 mg/L,相关系数均大于0.995,检出限(LOD)为0.05-0.40 mg/kg。在3种加标水平下的平均加标回收率为85.1%~98.5%,相对标准偏差为1.2%-5.3%(n=6)。该方法准确、灵敏度高、重现性好,适用于加工肉制品中11种防腐剂和抗氧化剂的测定。 To establish a high-performance liquid chromatography with diode array detector(HPLC-DAD)method separated with gradient elution for the determination of 11 preservatives and antioxidants in processed meat, which based on a kind of pretreatment method by solid-phase extraction for the determination of processed meat with complicated matrices. The sampleswere separated on HPLC-DAD system with aXtimate C18 column and detected by converting UV-Vis wavelength,which quantified by standard curve method. The mobile phases were methanol and 0.02 mol/L ammonium acetate in gradient elution.thecalibration curves were linear over the concentration range of 0-10.0 mg/L with the correlation coefficients were allabove 0.995.The detection limits(3S/N) of this method were 0.05-0.40 mg/kg. The average recovery rates of 3 concentration levels in processed meat werebetween 85.1% and 98.5% with the relative standard deviations of spiked samples varied from 1.2% to 5.3%(n=6). The method is accurate,sensitive and reproducible,and is suitable for the determination of 11 antiseptics and antioxidants.
