葛根素在内毒素性急性肺损伤中抗炎作用的研究 被引量:2

摘要 目的探讨葛根素在内毒素性急性肺损伤中的抗炎作用。方法将成年SD大鼠随机分为对照组、内毒素模型组(LPS组)和葛根素组(Pu组)。检测大鼠肺组织的湿/干重比值(W/D)和支气管肺泡灌洗液(BALF)中总蛋白的含量,以反映肺组织水肿的程度。在给予脂多糖(LPS)后1h、3h和6h3个时间点,检测血清和BALF中肿瘤坏死因子α(TNF-α)和IL-6的变化。同时检测肺组织髓过氧化物酶(MPO)的活性变化和BALF中总细胞、中性粒细胞和巨噬细胞的计数,以反映肺内炎症反应的程度。结果①与对照组相比,LPS组肺组织W/D比值和BALF中总蛋白的含量明显升高(P<0.05)。而给予葛根素后,与LPS组相比,Pu组W/D比值和BALF中总蛋白的含量均显著降低(P<0.05).②与对照组相比,给予LPS后,大鼠血清和BALF中TNF-a和IL-6的含量明显升高(P<0.05);肺组织MPO的活性和BALF中总细胞、中性粒细胞和巨噬细胞的计数也明显增高(P<0.05)。而给予葛根素后,与LPS组相比,Pu组TNF-α和IL-6的含量和MPO活性均显著降低,BALF中总细胞、中性粒细胞和巨噬细胞的计数也明显减少(P<0.05)。结论葛根素可以显著抑制LPS诱导的炎症反应,对内毒素性急性肺损伤有明显的保护作用。 Objective To investigate the anti-inflammatory effects of puerarin on lipopolysaccharide(LPS)-induced acute lung injury.Methods Adult SD rats were randomly divided into control group,LPS group and Pu group.The wet/dry weight(W/D)ratio and the total protein content in bronchoalveolar lavage fluid(BALF)were measured to reflect the degree of pulmonary edema.The concentrations of TNF-αand IL-6 in serum and BALF were detected at 1 h,3 h and 6 h after LPS administration.Meanwhile,the activity of myeloperoxidase(MPO)and the numbers of inflammatory cells in BALF were detected to reflect the degree of inflammation.Results①Compared with control group,the W/D ratio and the total protein content in BALF significantly increased after LPS administration(P<0.05).However,puerarin administration obviously decreased the W/D ratio and the total protein content in BALF compared with LPS group(P<0.05).(2)Compared with control group,the concentrations of TNF-αand IL-6 in serum and BALF were significantly increased after LPS administration(P<0.05).Meanwhile,the activity of MPO in lung tissues was significantly increased,and the numbers of total cells,neutrophils and macrophages in BALF were obviously increased(P<0.05).However,puerarin administration obviously decreased the concentrations of TNF-αand IL-6,the activity of MPO and the numbers of total cells,neutrophils and macrophages in BALF(P<0.05).Conclusions Puerarin can significantly inhibit the LPS-induced inflammatory reaction and has obvious protective effects on LPS-induced acute lung injury.
