维吾尔族和汉族妇女宫颈癌与宫颈CINⅡ~Ⅲ级组织中miR-145的表达与意义 被引量:3

Expression and significance of miRNA-145 in cervical carcinoma and cervical intraepithelial neoplasiaⅡ-Ⅲ level tissues of Uygur and Han women

摘要 目的检测miR-145在新疆维吾尔族和汉族妇女宫颈癌与宫颈上皮内瘤变(CIN)Ⅱ~Ⅲ级组织中的表达情况,探讨其与宫颈癌病理特征的关系及意义。方法采用实时荧光定量qRT-PCR技术检测维吾尔族和汉族妇女宫颈石蜡组织标本(宫颈癌50例、CINⅡ~Ⅲ级48例、正常宫颈22例)中miR-145的表达,分析其在维吾尔族和汉族妇女不同宫颈病变中的表达差异及其与宫颈癌常见病理特征的关系,绘制受试者工作曲线(ROC)分析miR-145作为宫颈癌肿瘤标记物的诊断价值。结果 miR-145在维吾尔族(χ~2=12.531)和汉族(χ~2=13.619)妇女宫颈癌组织中的表达均显著低于CINⅡ~Ⅲ级组织和正常宫颈组织,差异有统计学意义(均P<0.05),同类宫颈病变组织维吾尔族和汉族妇女的表达差异无统计学意义(均P>0.05);miR-145的表达量与宫颈癌组织分化程度(Z=-2.816)、FIGO分期(χ~2=13.513)、淋巴结转移(Z=-2.210)表达有差异(均P<0.05),与病人年龄(Z=-0.586)、肿瘤直径(Z=-0.799)、病理类型(Z=-0.986)、肌层浸润(Z=-1.134)均无相关(均P>0.05);ROC曲线分析miR-145区分宫颈癌与正常者的曲线下面积为0.887(95%CI:0.811~0.964,P=0.000),灵敏度81.8%,特异度76.0%。结论 miR-145参与宫颈癌的发生发展,在宫颈癌组织中表达明显下调,且无民族表达差异,可能成为宫颈癌诊断、靶向治疗及病情预测的新型分子标记。 Objective To detect miR-145 expression in cervical carcinoma and cervical intraepithelial neoplasia(CIN)tissues at grade Ⅱ-Ⅲ in Uygur and Han women of Xinjiang, and explore the relationship between the miR-145 expression and clinicopathologic features of cervical carcinoma and its significance. Methods Quantitative real-time PCR was taken to detect miR-145 expression in the specimens of cervical paraffin embedded tissue(50 specimens of cervical carcinoma, 48 specimens of CIN gradeⅡ-Ⅲ and 22 normal specimens) of Uygur and Han women. The differences in miR-145 expression among different cervical diseases and the relationship between the expression and clinicopathologic features of cervical carcinoma were analyzed. The ROC curve was drawn, and the diagnostic value of miR-145 as biomarker of cervical carcinoma was analyzed. Results The miR-145 expression was significantly lower in cervical carcinoma than in CIN tissues of grade Ⅱ-Ⅲ and normal cervical tissues of Uygur(χ2=12.531) and Han women(χ2=13.619) with statistical difference(P<0.05). There were no statistical differences(all P>0.05) in the miR-145 expression in the same kinds of cervical lesions between Uygur and Han women. The expression level of miR-145 was related with(all P<0.05)differentiation degree of the cervical tissues(Z=-2.816), FIGO stage(χ2=13.513) and lymph node metastasis(Z=-2.210);but without(all P>0.05) patient age(Z=-0.586), tumor diameter(Z=-0.799), pathological types(Z=-0.986) and myometrial invasion(Z=-1.134). The area under ROC distinguishing cervical carcinoma and the normal by miR-145 was 0.887(95%CI:0.811-0.964, P=0.000), with the sensitivity of 81.8% and specificity of 76.0%. Conclusions The miR-145 involves in the oncogenesis and development of cervical carcinoma and expresses down-regulation in the tissues of cervical carcinoma without ethnic difference. It may become potential biomarkers for diagnosis, targeted therapy and disease prognosis of cervical carcinoma.
