环氧合酶-2对酸和胆盐诱导的人鳞状细胞凋亡的影响及机制

摘要 目的探讨环氧合酶-2(COX-2)对酸和胆盐诱导的人鳞状细胞凋亡的影响及机制。方法体外培养人食管鳞状细胞,瞬时转染COX-2 SiRNA载体,48 h后经酸、胆盐及二者混合物处理细胞株,流式细胞术检测细胞凋亡,同时运用蛋白印迹技术检测COX-2、核转录因子-κB(NF-κB)及尾型同源盒转录因子-2(CDX-2)表达。结果酸、胆盐及二者混合物均诱导细胞凋亡,酸和胆盐的混合物作用最强,基因沉默COX-2后细胞凋亡增加,伴随NF-κB及CDX-2表达降低。结论基因沉默COX-2可以增加酸和胆盐诱导的人鳞状细胞凋亡,通过COX-2激活NF-κB信号通路及促进CDX-2表达可能是其机制之一。 Objective To investigate the effect of COX-2 on apoptosis of human esophageal squamous epithelium induced by acid and bile salts and its possible molecular mechanism.Methods Human esophageal squamous epitheliums were cultured in vitro and transfected instantaneously with COX-2 SiRNA vector.After 48 hours,cell lines were treated with acid,bile salts and a mixture of both,and cell apoptosis was detected by flow cytometry.Meanwhile,expressions of COX-2,NF-κB and CDX-2 were detected by western blot.Results Apoptosis was induced by acid,bile salts and their mixtures,and acid and bile salts mixtures have the strongest effect.Apoptosis was increased after gene silencing COX-2,accompanied by decreased expression of NF-κB and CDX-2.Conclusion Gene silencing COX-2 can increase the apoptosis of human esophageal squamous epithelium induced by acid and bile salts,and the activation of NF-κB and CDX-2 signaling pathway may be one of its mechanisms.
