骨髓干细胞通过多种机制促进缺血再灌注小鼠肾脏修复 被引量:2

摘要 目的通过肾脏动脉夹闭的方法制成急性肾损伤模型,并通过相关指标的检测证实干细胞的保护作用及相关机制。方法通过提取小鼠胫骨骨髓采取贴壁培养的方法体外培养扩增骨髓间充质干细胞,为了进一步在体内追踪干细胞,在将干细胞注入模型鼠体内之前,将干细胞用绿色荧光染料(CMFDA)染色,并通过尾静脉注射的方法注入到急性缺血再灌注小鼠模型鼠体内,在不同时间点通过检测肾脏功能及体质量,并通过肾脏病理HE染色及免疫荧光染色的方法观察增生细胞及管周毛细血管等指标比较骨髓间充质干细胞的肾脏保护作用。结果经流式细胞仪检测、免疫荧光及光镜证实培养的细胞具有典型的干细胞形态及细胞表面分子标记物,注入小鼠体内的干细胞可以特异性被招募到受损肾脏,注入干细胞后明显促进急性缺血再灌注小鼠模型的肾脏功能、体质量及生存率,促进细胞再生并减少肾脏管周毛细血管的丢失,促进肾脏病理的改善。结论骨髓间充质干细胞促进急性缺血再灌注肾脏损伤的修复。 Objective Acute renal failure(ARF)is a potentially devastating problem in clinical medi cine,therapeutic advances have been limited because of the complexity of kidney.More and more studies have shown that stem cells provide the beneficial therapy but the effect and the mechanism still await investigation.To investigate the role of the mesenchymal stem cells(MSCs)after ischemia reperfusion(I/R),we induced I/R modal and injected MSCs via vein tail.Methods MSCs were isolated and cultured from bone marrow of male BALB/C mice,then expand to at least tenth passage used to transplant to the I/R modal.Renal function,histological changestested by HE staining,cells proliferation and peritubularcapillaries(PTC)measured by immunofluorescent staining were examined at 3,5,7 day after I/R.Results MSCs transplantation improved the renal function,reduced mortality and markedly lowered the severityof histological changes.MSCs transplantation also promoted repair of the kidney microvas culature,attenuated kidney PTCloss and increased the proliferation of parachymal cells.Conclusions Inconclusion,the data shows that MSCstransplantation improved the kidney repair after I/R injury by increasing regeneration of PTC and cells proliferation.
