改良丙酮沉淀法用于天花粉蛋白的提取

摘要 随着天花粉蛋白(TCS)在临床性研究的不断进展,天花粉蛋白的生产工艺也越来越受到人们的重视。分别利用硫酸铵盐析法和改良丙酮沉淀法对天花粉蛋白的提取和工艺进行了比较分析,硫酸铵盐析法提取天花粉蛋白过程中操作复杂,提取率低,仅为0. 464‰;改良丙酮沉淀法操作简便,可短时间多次提取天花粉样品,提取率为1. 152‰,提取效果明显优于传统的硫酸铵分级沉淀法。由此表明改良丙酮沉淀法更适合天花粉蛋白质的研究。 With the development of trichosanthin in clinical research,more and more attentions have been paid to the technology of protein production.The extraction process and technology of TCS were compared and analyzed by ammonium sulfate precipitation and modified acetone precipitation respectively.And the extraction efficiency of the two methods was compared.Ammonium sulfate salt chromatography was complex in the process of extracting celestial pollen protein,and the extraction rate was low,only 0.464‰.The improved acetone precipitation method was simple to operate,and it can extract pollen samples several times in a short period of time.The extraction rate was 1.152‰,and the extraction effect was significantly better than the traditional ammonium sulfate grading precipitation method.The results showed that the improved acetone precipitation method was more suitable for the study of trichosanthin.
