摘要 目的建立HPLC法测定蒙药地格达-4汤中秦皮乙素的含量。方法在单因素实验基础上,采用Box-Behnken响应面法优选出地格达-4汤中秦皮乙素的提取工艺,采用HPLC法测定地格达-4汤中秦皮乙素的含量。色谱条件:πNAP cosmosil C_(18)色谱柱(250 mm×4.6 mm,5μm);以甲醇-1 mL·L^(-1)磷酸(21∶79)为流动相;流速为1.0 mL·min^(-1);检测波长为345 nm;柱温为30℃。结果秦皮乙素质量浓度在3.47~888.00μg·mL^(-1)范围内线性关系良好(r=0.999 8);平均加样回收率为96.0%。结论该方法简便、准确,可有效控制地格达-4汤的质量。 Objective To establish an HPLC method for the determination of aesculetin in Mongolian medicine Digeda-4 Decoction. Methods Based on the results of single factor test,the Box-Behnken design-response surface method was used to optimize the extraction process of aesculetin from Digeda-4 Decoction.The content of aesculetin in Digeda-4 Decoction was determined by HPLC.The chromatographic conditions were as follows:a π NAP cosmosil C18 (250 mm×4.6 mm,5 μm) column was used,the mobile phase was a mixture of methanol-1 mL·L^-1 phosphoric acid (21 ∶79),the flow rate was1.0 mL·min^-1 ,the detection wavelength was 345 nm,and the column temperature was 30 ℃. Results The method showed good linear relationship within the range of 3.47-888.00 μg· mL -1 for aesculetin ( r =0.999 8),and the average recovery was 96.0%. Conclusion The method is simple and accurate,and can effectively control the quality of Digeda-4 Decoction.
