百合地黄汤不同萃取部位的镇静催眠活性研究 被引量:10

摘要 目的以果蝇为模式生物筛选百合地黄汤提取物镇静催眠的有效部位,并基于单胺类神经递质的水平对各有效部位的镇静催眠作用进行研究。方法百合地黄汤提取物依次用石油醚、二氯甲烷、乙酸乙酯和水饱和正丁醇萃取,分离得不同萃取部位,以果蝇失眠模型筛选镇静催眠有效部位,并研究各有效部位对果蝇脑部单胺类神经递质的影响。结果与空白组相比,模型组果蝇总睡眠时间显著缩短(P<0.01);与模型组相比,百合地黄汤全方低、中、高剂量组果蝇总睡眠时间明显延长(P<0.05),石油醚部位低剂量组、二氯甲烷部位中剂量组、乙酸乙酯部位中剂量组以及水饱和正丁醇部位低剂量组果蝇总睡眠时间明显延长(P<0.05);各有效部位组果蝇脑部多巴胺(DA)及其代谢物高香草酸(HVA)含量降低,5-羟色胺(5-HT)及其代谢物5-羟吲哚乙酸(5-HIAA)含量升高。结论百合地黄汤提取物的石油醚、二氯甲烷、乙酸乙酯以及水饱和正丁醇部位均有镇静催眠作用,推测各有效部位的镇静催眠作用可能与调节脑部单胺类神经递质水平相关。 Objective To study the sedative and hypnotic activity of different solvent fractions of Baihe Dihuang Decoction by using Drosophila as a model organism.The effective sites were further evaluated though the expression level of monoamine neurotransmitters in Drosophila . Methods Baihe Dihuang Decoction was extracted with petroleum ether,dichloromethane,ethyl acetate and water saturated n -butanol successively to obtain the different polarity extraction parts.The sedative-hypnotic effective sites were screened by Drosophila insomnia model,and the effective sites were further evaluated through monitoring the expression level of monoamine neurotransmitters in Drosophila brain. Results Compared with the blank group,the sleep time of Drosophila in the model group was significantly shortened ( P <0.01),indicating the model was established successfully.Compared with the model group,the sleep time of Drosophila was obviously prolonged in the Baihe Dihuang Decoction low,middle and high group ( P <0.05),and sleep time of Drosophila was obviously prolonged in the low-dose group of petroleum ether extract,medium-dose group of dichloromethane extract,medium- dose group of ethyl acetate extract and low-dose group of water- saturated n-butanol extract ( P <0.05).The contents of dopamine (DA) and homovanillic acid (HVA) in the brain of Drosophila were decreased in previously screened functional groups,while the contents of 5- hydroxytryptamine (5-HT) and 5-hydroxyindole acetic acid (5-HIAA) were increased. Conclusion Petroleum ether,dichloromethane,ethyl acetate and water saturated n -butanol extracts from Baihe Dihuang Decoction have the sedative-hypnotic effects.Their sedative-hypnotic effects may be related to the regulation of the expression level of monoamine neurotransmitters in brain.
