ELISA法与免疫膜条法检测抗MDA5抗体、抗TIF1γ抗体的比对分析 被引量:4

摘要 目的比较酶联免疫吸附试验(ELISA)和免疫膜条法检测抗黑色素瘤分化相关基因5(MDA5)抗体和抗转录中介因子1γ(TIF1γ)抗体结果的差异性。方法选取日本MBL公司生产的抗MDA5抗体和抗TIF1γ抗体试剂盒(ELISA法)与德国欧蒙公司抗肌炎抗体谱IgG检测试剂盒(免疫膜条法),同时检测180例皮肌炎血清样本中的抗MDA5抗体和抗TIF1γ抗体,比较这两种方法的差异性。结果两种方法检测到的两种抗体阳性率,差异均无统计学意义(P>0.05)。两种方法检测结果的一致性有统计学意义(P<0.001),但一致性一般(抗MDA5抗体:Kappa值=0.660;抗TIF1γ抗体:Kappa值=0.642)。结论 ELISA法和免疫膜条法检测抗MDA5抗体和抗TIF1γ抗体的一致性程度一般。免疫膜条法有待进一步改善,以提高检测的准确性。 Objective To compare the difference between enzyme-linked immunosorbent assay (ELISA) and a line blot assay in detecting anti-melanoma differentiation-associated protein 5 (MDA5) antibody and anti-transcriptional intermediary factor 1γ(TIF1γ) antibody. Methods The anti-MDA5 antibody and anti-TIF1γ antibody kit (ELISA method) produced by MBL Company of Japan and the anti-myositis antibody spectrum IgG detection kit (line blot method) produced by Omon Company of Germany were selected to detect anti-MDA5 antibody and anti-TIF1γ antibody in 180 serum samples of dermatomyositis,and the differences between the two methods were compared. Results There was no significant difference in the positive rates between ELISA and line blot assays ( P >0.05).The consistency of the results of the two methods was statistically significant ( P <0.001),which was moderate for anti-MDA5 antibody ( Kappa :0.660) and anti-TIF1γ antibody ( Kappa :0.642). Conclusion A moderate consistency between ELISA and line blot was found for anti-MDA5 antibody and anti-TIF1γ antibody.The line blot assay should be improved to enhance its accuracy.
