谷氨酸转运体对心肌肥大的影响作用研究

摘要 目的探索谷氨酸转运体(GT)对心肌肥大的影响。方法选取50只7周龄的野生型雌性Balb/c小鼠(A组)和50只7周龄的ISO诱导心肌肥大型雌性Balb/c小鼠(B组),饲养1个月以后,取小鼠心脏,采用直接测量和心脏超声的方法测量心重/体重、心肌细胞体积、心室壁厚度及各心腔的具体指标水平。取小鼠心肌原代细胞培养,用实时荧光定量聚合酶链反应(qPCR)和蛋白免疫印迹法(Western blotting)等方法检测GT各基因在A、B组小鼠心肌细胞中的表达情况。结果 EAAT3基因在正常小鼠中表达更高。结论心肌肥大时EAAT3的表达降低。 Objective To explore the effect of glutamate transporter (GT) on cardiac hypertrophy. Methods Fifty 7-week-old wild-type female Balb/c mice (group A) and fifty 7-week-old ISO-induced cardiac hypertrophic large female Balb/c mice (group B) were selected.After 1 month of feeding,the hearts of the mice were removed,and the heart weight/body weight,myocardial cell volume,ventricular wall thickness,and specific index levels of each heart chamber were measured by direct measurement and cardiac ultrasound.Primary mouse cardiomyocytes were cultured,and the expression of GT genes in cardiomyocytes of mice A and B was detected by real-time fluorescent quantitative polymerase chain reaction (qPCR) and Western blotting. Results The EAAT3 gene was expressed more in normal mice. Conclusion The expression of EAAT3 is decreased in cardiac hypertrophy.
