子宫内膜衰老退行性变动物模型的建立

Establishment of rat model of electrocoagulation-injured endometrium

摘要 目的建立稳定的大鼠子宫内膜(电凝损伤)衰老退行性变模型。方法将单极电针插入大鼠的一侧子宫腔,以1V、3V、10V的不同电压制造损伤。设置对侧子宫作为对照。比较损伤7天后不同侧子宫进行石蜡切片HE染色后,相应子宫内膜厚度及相同高倍视野下子宫内膜腺体的数目。结果接受3V电压损伤的子宫,术后7天其内膜厚度为1.44±0.56mm,与未接受电压损伤的子宫内膜厚度4.44±1.13mm相比,差异有统计学意义。在高倍视野下计数子宫内膜腺体数目,3V电压损伤侧腺体个数为1.44±1.59,未接受电压损伤侧的腺体个数为5.88±1.90,差异有统计学意义。结论以3V电压刺激子宫内膜所造成的损伤可以模拟子宫内膜损伤后的形态学改变,适用于大鼠子宫内膜(电凝损伤)衰老退行性变动物模型的研究。 Objective To establish the rat model of electrocoagulation-injured endometrium.Methods Unipolar electric acupuncture was applied to make injury on one side of uterus with the voltage of 1V,3V and 10V,with the other side uterus as control.7 days after injury,the endometrial histomorphology was observed by microscope and number of gland was calculated.Results The average endometrium thickness of 3V electrocoagulation injured group is 1.44±0.56,while in non-electrocoagulation injured side is 4.44±1.13.The difference is significant.Also,the number of gland observed in high power microscope of 3V injured side is 1.44±1.59,which is significantly lower than 5.88±1.90 in non-electrocoagulation injured side.Conclusion The electrocoagulation injury of 3 voltage can give rise to morphology changes similar to clinical endometrial injury and can be applied to animal research of rat endometrial injury.
