矮凤梨组织培养快速繁殖技术研究 被引量:2

摘要 本试验用矮凤梨(Ananas comosus var. nanus)的吸芽作为外植体,研究了在光照培养和黑暗培养条件下不同激素配比对矮凤梨愈伤组织增殖率的影响;同时也研究了移栽时株高和栽培基质对矮凤梨组培苗的成活率和生长的影响。结果表明:在光照条件下矮凤梨愈伤组织的增殖率显著高于其在黑暗条件下的增殖率。光照培养下矮凤梨愈伤组织最佳的增殖培养基为MS+6-BA 3.0 mg·L^(-1)+NAA 3.0 mg·L^(-1);黑暗培养条件下则为MS+6-BA 3.0 mg·L^(-1)+NAA 2.0 mg·L^(-1)。定植时的株高和栽培基质对矮凤梨组培苗成活率无显著影响,组培苗在株高为5 cm≤x<7 cm时进行定植、栽培且在泥炭土和蛭石比为3∶1的基质中综合生长状况较佳。 In this experiment,suckers of Dwarf pineapple(Ananas comosus var.nanus)were used as explants to study the effects of different hormone combinations on callus proliferation rate under light and dark culture condition.The results showed that the callus proliferation rate of Ananas nanus under the light conditions was significantly higher than that under the dark conditions.The best proliferation medium of callus was MS+6-BA 3.0 mg·L^-1+NAA 3.0 mg·L^-1 under the light while the best proliferation medium of callus was MS+6-BA 1.0 mg·L^-1+NAA 3.0 mg·L^-1 under dark condition.In addition,the effects of plant height and substrate in transplanting on survival rate and growing situation of plantlets were studied.The results showed that plant height and substrate had no significant effects on the survival rate of the plantlets.When the tissue culture seedlings were planted at the height between 5~7 cm,and were cultivated in a matrix with a peat soil and vermiculite ratio of 3∶1,the seedlings showed better than others.
