通城虎醇提物对大鼠的肾毒性研究

摘要 目的研究通城虎醇提物对单肾切除大鼠的肾毒性。方法雌雄各半的SD大鼠50只,随机分为空白对照组、模型对照组、通城虎醇提物给药组(分为高剂量组、中剂量组、低剂量组),每组10只。除空白对照组以外,对其余各组大鼠进行右肾切除手术。术后第5天开始连续灌胃给药14 d,空白对照组和模型对照组给予同等体积的蒸馏水。绘制体重变化曲线,测算部分脏器系数,测定尿NAG酶、BUN、CREA、UA生化指标。结果与空白对照组相比,通城虎醇提物给药组的大鼠体重减轻(P<0.05);NAG酶均明显升高(P<0.05);高剂量组BUN、CREA、UA等指标明显升高(P<0.05);中剂量组CREA、UA指标明显升高(P<0.05),低剂量组指标CREA明显升高(P<0.05)。病理学检查显示,通城虎对大鼠的肝脏无明显损伤作用,而对肾脏具有明显损伤毒性。结论通城虎对单肾切除的大鼠肾脏具有明显的毒性。 Objective To study the nephrotoxicity of ethanol extract of Aristolochia fordiana Hemsl on rats with single nephrectomy.Methods Fifty male and female SD rats were randomly divided into control group,model control group and Aristolochia fordiana Hemsl alcohol extract group (divided into high dose group,middle dose group and low dose group),10 rats per group.Except for the blank control group,the other groups of rats underwent right nephrectomy.On the 5th day after the operation,continuous intragastric administration was started for 14 days,and the blank control group and the model control group were given the same volume of water.The weight change curve was drawn,some organ coefficients were measured,and the biochemical indicators of urinary NAG enzyme,BUN,CREA,and UA were determined.Results Compared with the blank control group,the weight of the rats in the drug-administered group was reduced ( P <0.05).The BUN,CREA,UA and other indicators in the high-dose group were significantly increased ( P <0.05).The CREA and UA indexes in the middle-dose group were significantly increased ( P <0.05),and the CREA in the low-dose group was significantly higher ( P <0.05).Pathological examination showed that there were no observed in Aristolochia fordiana Hemsl damage to the liver of rats,but had obvious damage toxicity to the kidney.Conclusion Aristolochia fordiana Hemsl is observed the toxicity to the kidney of single nephrectomy rats.
