HPLC法对斑花黄堇中普托品的含量测定

摘要 目的建立藏药斑花黄堇中普托品含量的测定方法。方法采用高效液相色谱法,选用C18柱(型号:Eclipse XDB,250 mm×4.6 mm,5μm)为色谱柱,选择甲醇-0.1mol/L乙酸铵溶液-磷酸-氨水体系为流动相,流速为1.0 mL/min,检测波长为285.16 nm,柱温30℃,以外标法检测。结果供试品溶液色谱中,普托品在0.05 mg/m L~0.80 mg/m L范围内与峰面积呈良好线性关系,回归方程为Y=12 109X+76.19,平均回收率为103.42%,RSD=2.20%(n=9)。结论该方法样品分离度佳、灵敏度高、重现性好,结果准确可靠,适合于斑花黄堇中普托品的测定。 Objective To establish an HPLC method for determination of protopine in Corydalis conspersa Maxim.Methods The Eclipse XDB-C18(200 mm×4.6 mm,5μm)column was used,the mobile phase consisted methanol-0.1 mol/L Ammonium acetate-Phosphoric-Ammonia water,the flow rate was 1.0 mL/min and the detection wavelength was 285.16 nm.Results The established HPLC method had good linearity and precision within the range of 0.05 mg/mL^0.80 mg/mL of protopine.regression equation:Y=12109 x+76.19(r=0.9997).Average recovery is 103.42%,RSD=2.20%(n=9).Conclusion The method is good in separation,sensitivity and reproducibility.The result is accurate and reliable,and can be applied to the determination of the content of putropin in Corydalis Conspersa Mazim.
