摘要
目的通过生物信息学分析筛选出与胰腺导管腺癌(pancreatic ductal adenocarcinoma,PDAC)患者生存相关的长链非编码RNA(lncRNA),并预测其靶基因及相关信号通路。方法通过肿瘤基因组图谱(The Cancer Genome Atlas,TCGA)数据库下载PDAC及癌旁组织的表达谱数据及相应的临床资料,并对这些数据进行生存分析,然后通过共表达分析及基因富集分析找到与差异lncRNA表达相关的mRNA和信号通路。最后通过细胞实验验证AC015660.1、CASC8在PDAC细胞中的表达。结果通过TCGA筛选出4个在PDAC中差异表达的lncRNA,其中AC015660.1和CASC8表达上调,对其进行共表达分析,分别预测出2个靶基因mRNA;对进行单基因GSEA分析,预测出8条CASC8可能的作用通路。细胞实验显示,AC015660.1和CASC8均在PDAC细胞中高表达。结论通过生物信息学分析筛选出与PDAC生存相关的lncRNA,为胰腺癌的预后评估提供了潜在靶点。
Objective To screen long-chain non-coding RNA(lncRNA) related to the survival of pancreatic ductal adenocarcinoma(PDAC) patients by bioinformatics analysis, and to predict its target genes and expression-related pathways. Methods The expression profile data and corresponding clinical data of PDAC and adjacent tissues were downloaded from the Cancer Genome Atlas(TCGA) database, and the data were analyzed for survival and then analyzed by co-expression. Gene enrichment analysis found mRNA and signaling pathways associated with differential lncRNA expression.Results Based on the survival analysis of FC>2 and P<0.05, it was found that lncRNA AC015660.1, AC108752.1, NFIA-AS2 and CASC8 were closely related to the prognosis of PDAC.Conclusion Screening for lncRNA associated with PDAC survival by bioinformatics analysis provides a potential target for prognostic evaluation of pancreatic cancer.
