氨氮急性攻毒及去毒后棘胸蛙蝌蚪肝脏和尾部皮肤相关功能酶活力的变化特征 被引量:2

Study on Changes of Related Enzyme Activities in Liver and Tail Skin of Paa spinosa Tadpole after Ammonia Nitrogen Acute Challenge and Detoxification

摘要 以体长1.932±0.204 cm、体质量1.386±0.055 g的棘胸蛙蝌蚪为实验动物,在水温24±0.2℃、DO7.30±0.01 mg·L^(-1)、pH 7.30±0.01条件下,以96 h为氨氮急性攻毒时长,12 d为去毒恢复时长,采用静水停食法,研究了氨氮急性攻毒及去毒恢复阶段棘胸蛙蝌蚪肝脏SOD、CAT、GSH及尾部皮肤SOD、CAT、ATP酶活力的变化特征。结果表明:(1)急性攻毒阶段,所涉酶类中除尾部皮肤ATP酶活力随氨氮攻毒质量浓度增加呈显著下降的趋势(P<0.05)外,其余酶活力均随氨氮质量浓度增加呈先升后降趋势,其中SOD酶活力的峰值出现于氨氮质量浓度2.45 mg·L^(-1)实验组(P<0.05),CAT和GSH酶活力峰值均出现于氨氮质量浓度14.70 mg·L^(-1)实验组(P<0.05);(2)去毒恢复阶段,所涉酶类酶活力随原氨氮急性攻毒质量浓度增加均呈先升后降的变动特征,且峰值均出现于原氨氮质量浓度17.15 mg·L^(-1)实验组(P<0.05)。 Paa spinosa tadpole with body length 1.932±0.204 cm and body mass 1.386±0.055 g was used as experimental animal at water temperature 24±0.2 °C, DO 7.30±0.01 mg·L^-1, pH 7.30±0.01, 96 h was the duration of acute challenge of ammonia nitrogen, and 12 d was the duration of detoxification recovery. Under the conditions of smooth water and feed deprivation, the changes on liver’s SOD, CAT, GSH activities and tail skin’s SOD, CAT, ATP activities of P. spinosa tadpole on the stage of ammonia-nitrogen acute attack and detoxification recovery were studied. The results showed that:(1)In the acute challenge stage, the ATPase activity of tail skin decreased significantly with the increment of ammonia nitrogen concentration(P<0.05), whilethe other enzymes activities increased first and then decreasedwith the increment of ammonia nitrogen concentration, among which the peak value of SOD activity appeared in experimental group whose ammonia nitrogen concentration was 2.45 mg·L^-1(P<0.05), and thepeak value of CAT and GSH activitiesboth were found in the group whose ammonia nitrogen concentration was14.70 mg·L^-1(P<0.05);(2) At the stage of detoxification recovery, all involvedenzymes activities increased first and then decreased with the increment of ammonia-nitrogen challengeconcentrations, and their peaks appeared in the group whose ammonia nitrogen concentration was 17.15 mg·L^-1(P<0.05).
