摘要
Fyn相关激酶(FRK)作为乳腺肿瘤激酶家族成员之一,是一种由505个氨基酸构成的非受体型酪氨酸激酶,其与肿瘤的发生发展密切相关。FRK主要通过磷酸化多种蛋白底物或与多种蛋白结合来行使其不同的细胞功能。目前,已知FRK能与人第10号染色体缺失的磷酸酶及张力蛋白同源基因、Rb及表皮生长因子受体等多种蛋白相互作用,并与细胞的增殖、迁移、侵袭及周期调控等密切相关。FRK的功能及表达均具有明确的细胞/组织特异性,其具体的表达水平及功能会因所在组织的不同而发生改变。未来,应深入研究确定FRK的新底物及结合蛋白,并明确FRK表达的调控机制。
Fyn-related kinase(FRK) as a member of breast tumor kinase family,is a non-receptor tyrosine kinase composed of 505 amino acids,which is closely related to the occurrence and development of tumors.FRK exerts its different cellular functions mainly by phosphorylating various protein substrates or binding to various proteins.At present,FRK has been known to interact with phosphates and tensin homologue deleted on chromosome ten gene,Rb and epidermal growth factor receptor proteins,and is closely related to cell proliferation,migration,invasion and cell cycle regulation.The function and expression of FRK have clear cell/tissue specificity,and the specific expression level and function of FRK change in different tissues.In the future,we should further study and identify new substrates and binding proteins of FRK,and clarify the regulatory mechanism of FRK expression.
