口腔鳞状细胞癌患者血浆lncRNAs差异表达谱的初步研究 被引量:2

摘要 目的:筛选与口腔鳞状细胞癌(oral squamous cell carcinoma,OSCC)相关的长链非编码RNA(long non-coding RNA,lncRNA),探讨OSCC相关lncRNAs在OSCC的发生、发展中的潜在作用机制。方法:选取OSCC TNM Ⅲ/Ⅳ期患者和健康对照组各3例,应用Arraystar人类lncRNA芯片(v4. 0)进行mRNA和lncRNA表达谱研究,对差异表达基因(differentially expressed genes,DEGs)进行GO和KEGG分析。结果:共有6606个lncRNAs表达有差异,其中上调3511个,下调3095个;有4196个mRNAs表达有差异,其中上调1766个,下调2430个。GO和KEGG分析显示DEGs功能涉及到肿瘤细胞的生长、分化、代谢、凋亡、信号传导等整个细胞周期过程,和肿瘤的增殖、迁移、侵袭等生物学行为密切相关。结论:筛选出了OSCC相关的差异表达lncRNAs,生物信息学分析提示它们参与了OSCC的发生、发展过程,是OSCC潜在的诊断和预后标志物,其作用机制还需要进一步验证。 Objective: To screen the long non-coding RNA (lncRNA) associated with oral squamous cell carcinoma (OSCC) and explore the potential role of OSCC-related lncRNAs in the occurrence and development of OSCC. Methods: Three cases of OSCC TNM III/IV patients and three cases of health control were selected, mRNA and lncRNA expression profiles were studied using Arraystar human lncRNA chip (v4 .0), and 14 lncRNAs were selected for qRT-PCR verification using chip homologous plasma samples. GO and KEGG analysis were performed on the differential expression genes (DEGs). Results: A total of 6606 differentially expressed lncRNAs were screened, of which 3511 were upregulated and 3095 were downregulated;A total of 4196 DEGs were screened, of which 1766 were upregulated and 2430 were downregulated. GO and KEGG analysis showed that the function of DEGs involves the entire cell cycle process, such as growth, differentiation, metabolism, apoptosis, and signal conduction of tumor cells. The involved pathways are closely related to the proliferation, migration, and invasion of tumors. Conclusion: Differential expression profile of plasma lncRNAs in patients with OSCC are identified. Bioinformatics analysis indicates that they participate in the occurrence and development of OSCC, which maybe potential diagnostic and prognostic markers of OSCC, and the mechanisms need to be further verified.
