糖尿病性膀胱病变与膀胱和腰骶背根神经节中神经生长因子水平长期下降的相关性 被引量:3

Experimental study on the correlation between diabetic cystopathy and long-term decrease of nerve growth factor levels in bladder and dorsal root ganglion of rats

摘要 目的探讨糖尿病性膀胱病变与膀胱和腰骶背根神经节中神经生长因子(nerve growth factor,NGF)表达水平的长期下降的相关性。方法采用随机数字发将30只雄性Wistar大鼠随机平均分为对照组与糖尿病组,每组15只。对照组受试大鼠不采取任何处理措施,糖尿病组受试大鼠构建糖尿病模型。造模1个月后,比较2组受试大鼠体质量、血糖、尿动力学指标及膀胱湿质量、残尿量,采用酶联免疫吸附测定(enzyme linked immunosorbent assay,ELISA)法检测2组受试大鼠膀胱和腰骶背根神经节中NGF蛋白表达水平,采用LOGSTIC多元回归分析糖尿病大鼠膀胱和腰骶背根神经节中NGF蛋白表达水平与尿动力学指标的相关性。结果造模后1个月,2组受试大鼠体重均明显高于造模前,糖尿病组受试大鼠体重明显低于对照组(P<0.05),糖尿病组受试大鼠血糖明显高于造模前及对照组(P<0.05),糖尿病组受试大鼠膀胱和腰骶背根神经节中NGF蛋白水平均明显低于造模前及对照组(P<0.05),糖尿病组受试大鼠膀胱湿质量、残尿量明显高于造模前及对照组(P<0.05),糖尿病组受试大鼠最大膀胱容量、顺应性明显高于造模前及对照组,最大膀胱压力明显低于造模前及对照组(P<0.05)。Logistic多因素回归分析结果显示,糖尿病大鼠膀胱和腰骶背根神经节中NGF蛋白表达水平长期下降与大鼠最大膀胱容量、顺应性呈显著负相关(P<0.05),与最大膀胱压力呈显著正相关(P<0.05)。结论膀胱和腰骶背根神经节中NGF表达水平的长期下降与糖尿病性膀胱病变发生相关。 Objective To investigate the correlation between diabetic cystopathy and long-term decrease of nerve growth factor (NGF) in bladder and dorsal root ganglion of rats. Methods Thirty male Wistar rats were randomly divided into control group ( n =15) and diabetes group ( n =15). The rats in control group were not treated,however,the rat models with diabetes mellitus were established in diabetes group.At 1 month after modeling, the body mass, blood glucose, urodynamic parameters, wet weight and residual urine volume of bladder were observed and compared between the two groups. ELISA was used to detect the expression of NGF protein in bladder and dorsal root ganglion in both groups. The correlation between the expression levels of NGF protein in bladder as well as dorsal root ganglion and urodynamics indexes was analyzed by Logistic multiple regression. Results At one month after modeling, the body weight in both groups was significantly higher than that before modeling, and the body weight in diabetes group was significantly lower than that in control group ( P < 0.05 ).The blood glucose levels in diabetes group were significantly higher than those before modeling and those in control group ( P <0.05), and the NGF protein levels in bladder and dorsal root ganglion of rats in diabetes group were significantly lower than those before modeling and those in control group ( P <0.05). The wet quality and residual urine volume in diabetes group were significantly lower than those before modeling and those in control group ( P <0.05).The maximum bladder volume and compliance in diabetes group were significantly higher than those before modeling and those in control group ( P <0.05), and the maximum bladder pressure in diabetes group was significantly lower than that before modeling and that in control group ( P <0.05).Logistic regression analysis showed that the long-term decrease of NGF protein expression in bladder and dorsal root ganglion of rats in diabetes group was closely negatively correlated with maximal bladder volume and compliance,however,which was positively correlated with maximal bladder pressure ( P <0.05). Conclusion The long-term decrease of NGF expression in bladder and dorsal root ganglion of rats is associated with the occurrence of diabetic bladder disease.
