隐丹参酮对宫颈鳞癌Siha细胞增殖及凋亡影响机制的研究 被引量:1

摘要 目的 观察隐丹参酮对宫颈鳞癌Siha细胞增殖及细胞凋亡的影响,探讨其作用机制。方法 四氮唑噻唑蓝法检测不同浓度隐丹参酮对Siha细胞活力影响;流式细胞术测定Siha细胞增殖周期的分布;Westernblot法检测Siha细胞中E6、P53、P21蛋白的表达。结果 不同浓度隐丹参酮(0.1、1、5、10、100μmol·L^-1)对Siha细胞的细胞毒性有明显剂量和时间依赖性;隐丹参酮可使Siha细胞的细胞周期构成发生明显的变化,并诱导细胞凋亡;Westernblot法结果表明,隐丹参酮作用于Siha细胞后E6蛋白表达逐渐降低,P53和P21的蛋白表达逐渐升高。结论 隐丹参酮对宫颈鳞癌Siha细胞有较强的细胞毒性,这与其阻滞Siha细胞处于G0/G1期,并诱导细胞凋亡,使S期细胞比例降低;抑制E6蛋白表达,使P53功能恢复,从而起到杀伤肿瘤细胞的作用。 Objective To observe the effect of cryptotanshinone on the proliferation and apoptosis of cervical squamous cell carcinoma Siha cells and to explore its mechanism. Methods The effect of different concentrations of cryptotanshinone on the activity of Siha cells was detected by tetrazoli-thiazole blue method. Flow cytometry was used to determine the proliferation cycle of Siha cells. Western blot was used to detect the expression of E6, P53 and P21 proteins in cells. Results Different concentrations of cryptotanshinone (0.1, 1, 5, 10, 100 mol·L^-1 )had significant dose-dependent and time-dependent effects on the cytotoxicity of Siha cells. Cryptotanshinone could significantly change the cell cycle composition of Siha cells and induce the apoptosis. Western blot results showed that after the action of cryptotanshinone on Siha cells, the expression level of E6 protein was gradually decreased, while the expression levels of P53 and P21 protein was gradually increased. Conclusion Cryptotanshinone has strong cytotoxicity on cervical squamous cell carcinoma Siha cells. The mechanism of its action may be that the growth of Siha cells stagnates in G 0/G 1 phase and induces apoptosis, which reduces the proportion of cells in S phase. Inhibition of E6 protein expression, recovery of P53 function, and initiation of apoptosis play a role in killing tumor cells.
