摘要
目的探讨黄芪甲苷对缺氧缺血性脑损伤(hypoxic ischemic brain damage,HIBD)新生大鼠的保护作用及其机制。方法制备新生大鼠HIBD模型,随机分为假手术组、HIBD模型组和黄芪甲苷治疗组,观察黄芪甲苷大鼠对日常行为的影响,并对实验大鼠脑进行病理组织学观察,用Western blot检测大鼠脑NF-κB蛋白表达水平。结果 HIBD动物活动减少,部分动物出现躁动;透射电镜观察显示,假手术组神经元细胞内线粒体形态基本正常;HIBD模型组神经元线粒体肿胀,胞质内空泡形成;黄芪甲苷治疗组神经元细胞状态有所好转,细胞变性程度较HIBD模型组缓解;HE染色图片显示,假手术组大鼠海马细胞层次分明,HIBD模型组大鼠神经细胞变性、坏死,海马锥体细胞层次紊乱,黄芪甲苷治疗组神经细胞变性、坏死较HIBD模型组减轻;Western blot检测结果显示,HIBD模型组NF-κB蛋白水平明显升高,与假手术组比较差异有统计学意义(P<0.05),黄芪甲苷治疗后,NF-κB蛋白水平降低,与HIBD模型组比较差异有统计学意义(P<0.05)。结论黄芪甲苷对HIBD新生大鼠有一定改善作用。
Objective To investigate the protective effect and mechanism of astragaloside on hypoxic ischemic brain damage(HIBD)in newborn rats.Methods Neonatal rats HIBD model were constructed,rats were randomly divided into control group,the HIBD model group and astragaloside treatment group,daily behavior were observed,and the pathological and histological observation of experimental rats were done,protein expression level of NF-κB in rats’brain were detected by Western blot.Results The HIBD animal activity decreased,some animals appeared restless;Transmission electron microscopy showed that the morphology of mitochondria in neurons of control group were basically normal;neurons of HIBD model group had swelling of mitochondria and formation of vacuoles in cytoplasm;neurons shape in astragaloside treatment group were better than those of HIBD model group,and the degree of cells degeneration were alleviated;HE staining images showed that hippocampal cells rowed distinct in control group,nerve cells degeneration and necrosis in HIBD model group,and hippocampal py-ramidal cell level disorded,but nerve cell degeneration and necrosis in astragaloside treatment group were reduced than that in HIBD model group;Western blot test results showed that the NF-κB protein levels in HIBD model group was obviously higher,the difference was statistically significant compared with control group(P<0.05),after treatment of astragaloside,the NF-κB protein levels decreased,the difference was statistically significant compared with HIBD model group(P<0.05).Conclusion Astragaloside have certain effect in newborn rats with HIBD.
