摘要
目的探讨钙敏感受体(CaSR)对氧化型低密度脂蛋白(OxLDL)诱导的大鼠主动脉平滑肌细胞(A7r5细胞)迁移的影响及分子机制。方法 OxLDL(40μg/mL)处理A7r5细胞24 h诱导细胞迁移;应用细胞划痕实验(伤口愈合实验)检测细胞迁移;应用Western blot检测细胞CaSR和MMP-2蛋白表达。结果与对照组相比,OxLDL促进平滑肌细胞的迁移(P<0.05),增加MMP-2的表达(P<0.01);与对照组相比,OxLDL上调了平滑肌细胞的CaSR表达(P<0.05);与OxLDL组相比,使用NPS 2390(CaSR拮抗剂)预处理A7r5细胞抑制了OxLDL诱导的细胞迁移(P<0.05)及MMP-2的表达(P<0.01)。结论 CaSR可能通过影响MMP-2的表达参与OxLDL诱导的大鼠主动脉平滑肌细胞迁移过程。
Objective To investigate the effect of calcium-sensing receptor(CaSR)on oxidized-low density lipoprotein(OxLDL)-induced migration in rat aorta smooth muscle cells(A7r5 cells)and molecular mechanism.Methods OxLDL(40μg/mL)for 24 h induced migration in A7r5 cells.Wound healing assay was used to detect cell migration.The protein expression of CaSR and MMP-2 was detected by Western blot.Results Compared with control group,OxLDL promoted the migration of smooth muscle cells and increased the expression of MMP-2.OxLDL up-regulated the expression of CaSR in smooth muscle cells.NPS2390(CaSR antagonist)inhibited OxLDL-induced cell migration and MMP-2 expression.Conclusion CaSR may participate in OxLDL-induced migration by regulating the expression of MMP-2 in A7r5 cells.
