IGFBP-2增强内皮细胞血管形成作用的研究 被引量:1

摘要 目的研究胰岛素样生长因子结合蛋白-2(insulin-like growth factor binding protein-2,IGFBP-2)对内皮细胞血管形成作用的影响。方法在人脐静脉内皮细胞(human umbilical vein endothelial cells,HUVECs)中研究IGFBP-2过表达及敲减(knock-down)情况下,比较内皮祖细胞(endothelial progenitor cells,EPCs)-内皮细胞黏附及内皮细胞小管生成的情况。结果在HUVECs过表达IGFBP-2促进内皮祖细胞-内皮细胞黏附。相反,IGFBP-2敲减,抑制氧糖剥夺诱导的内皮祖细胞-内皮细胞黏附。IGFBP-2显著增强EPCs对HUVECs血管形成的作用。结论外源性IGFBP-2可促进血管新生,从而改善缺血状况。 Objective To study the effect of insulin-like growth factor binding protein-2(IGFBP-2)on the angiogenesis of endothelial cells.Methods Adhesion of endothelial progenitor cells(EPCs)to endothelial cells and angiogenesis of endothelial cells were compared with the overexpression and knock-down of IGFBP-2 in human umbilical vein endothelial cells(HUVECs).Results Overexpression of IGFBP-2 in HUVECs promoted EPC-endothelial adhesion.Conversely,knock-down of IGFBP-2 inhibited oxygen-glucose deprivation-induced EPCendothelial adhesion.IGFBP-2 significantly enhanced the effect of EPCs on vascular formation of HUVECs.Conclusion Exogenous administration of IGFBP-2 may facilitate neovascularization and improve treatment of ischemic conditions.
