甲状腺癌组织中PI3Kp85α、EZRIN和ARID1A表达检测及作用探讨 被引量:4

摘要 目的:通过对甲状腺癌及癌旁正常组织中磷脂酰肌醇3-激酶p85α亚单位(PI3Kp85α)、EZRIN和抑癌基因染色质重建复合物SWI/SNF亚基(AT rich interactive domain 1A:ARID1A)的mRNA和蛋白质的表达情况进行检测,探讨PI3Kp85α、EZRIN和ARID1A的相互之间的关联性,及三个检测指标与甲状腺癌病理分类因素的关系,来试图阐明PI3Kp85α、EZRIN和ARID1A的表达在甲状腺癌发生、侵袭及转移中的意义。方法:收集2017年1月-2018年5月忑枣庄市立医院乳腺甲状腺外科就诊的患者,就诊前无任何措施的治疗,经病理诊断的甲状腺癌组织114例及癌旁正常甲状腺组织43例。应用免疫组织化法和RT-PCR的方法检测甲状腺癌及癌务正常甲状腺组织中的PI3Kp85α、EZRIN和ARID1A的表达水平(实验结果采用SPSS19.0软件进行统计分析学分析处理)。结果:①与癌务正常组织相比较,PI3Kp85α、EZRIN在甲状腺癌中的阳性表达率及mRNA的表达显著升高(P<0.01),而ARID1A阳性表达率及mRNA的表达显著降低(P<0.01);②ARID1A mRNA的表达与甲状腺癌分化恶性程度呈负相关(P<0.05r=-0.689);PI3Kp85α、EZRIN与甲状腺癌的恶性程度呈正相关。(P<0.05 r=0.625)结论:PI3Kp85α、EZRIN、ARID1A的异常表达与甲状腺癌有较高的相关性,ARID1A的缺失或降低,能够促进PI3Kp85α和EZRIN蛋白的高表达,进而促进甲状腺癌的发生、发展和侵袭;联合检测该三项指标,有助于甲状腺癌对潜能侵袭转移的判断,从而为甲状腺癌的治疗和疗效评估,提供依据。 Objective: This study aims to reconstruct the complex SWI/SNF sub-unit (PI3Kp85α), EZRIN and tumor suppressor chromatin recombination complex (AT rich interactive domain 1A) in thyroid carcinoma and paracancerous nonnal tissues: The mRNA and protein expression of ARIDIA, the correlation between PI3Kp85α, EZRIN and ARIDIA, and the correlation w让h pathological classification factors were explored to clarify the significance of the expression of PI3Kp85α. Determine EZRIN and ARDDIA in the occurrence, invasion and metastasis of thyroid cancer. Methods: A total of 114 cases of thyroid cancer and 43 cases of paracancerous normal thyroid tissue were coDected from January 2017 to May 2018 in the department of breast and thyroid surgery of Zaozhuang city hospital without any treatment measures. Method of immune organized and RT 一PCR method for detection of thyroid carcinoma and normal thyroid tissue adjacent to carcinoma PI3Kp85α, EZKIN and ARIDIA expression level (experimental results analyzed by SPSS 19.0 software analysis processing) Results: 1. Compared with normal tissue adjacent to carcinoma, PI3Kp85α, EZRIN MRNA expression in thyroid carci noma was significantly increased (P<0.01), while the expression of ARIDIA was significantly reduced (P<0.01). 2. The expression of AKID1A MRNA was negatively correlated with the malignancy of thyroid carcinoma differentiation (P<0.05)-0.689);PI3Kp85α and EZRIN were positively correlated with the malignancy of thyroid cancer.(P<0.05 r =0.625). Conclusion: PI3Kp85a, EZRIN and ARIDIA in thyroid cancer, played an important role in invasion and metastasis. AKIDIA absent can promote high expression of PI3Kp85α and EZRIN protein, thus promote the occurrenee and development of thyroid carcinoma and invasive. The combined detection of the three indicators can help the judgment of potential invasion and metastasis from thyroid cancer prognosis, both for the treatment of thyroid cancer and curative effect evaluation.
