黄连素抗小鼠幽门螺杆菌感染作用的研究 被引量:1

Study on the anti-Helicobacter pylori infection effects of berberine in mice

摘要 目的观察黄连素协助治疗小鼠幽门螺杆菌(Helicobacter pylori,Hp)感染的效果。方法建立Hp感染小鼠模型,按不同抗Hp治疗方案分为3组:标准三联组给予抗Hp标准三联治疗方案(阿莫西林+克拉霉素+奥美拉唑),黄连素组在标准三联疗法基础上加用黄连素,铋剂组在标准三联疗法基础上加用胶体果胶铋。连续给药2周,停药4周后采用胃黏膜Giemsa染色法、快速尿素酶试验检测Hp根除情况。通过体外继代培养Hp,采用琼脂稀释法进行体外抗生素敏感试验,检测黄连素对Hp的最低抑菌浓度。结果标准三联组、黄连素组、铋剂组Hp根除率分别为55.0%(22/40)、77.5%(31/40)、95.0%(38/40),黄连素组根除率低于铋剂组,高于标准三联组,差异有统计学意义(P<0.05)。体外实验黄连素对Hp标准菌株最低抑菌浓度为12.5mg/ml。结论黄连素对Hp有一定的抑菌能力,黄连素联合标准三联疗法,可以提高Hp根除率。 Objective To observe the anti-Helicobacter pylori(Hp) infection effects of berberine in mice. Methods A young rats model of Hp infection was established, then the rats were divided into 3 groups according to different anti-Hp treatments.The standard group was given a standard triple therapy(amoxilin+ clasamycin+ omeprazole), the berberine group was added with berberine on the basis of the standard triple therapy;and the bismuth group was added with bismuth on the basis of the standard triple therapy.All the three groups received continuous treatment for 2 weeks.The eradication condition of Hp was detected by gastric mucosa Giemsa staining and rapid urinary enzyme test 4 weeks after discontinuation.In addition, the minimum antibacterial concentration of berberine was detected by using agar dilution method antibiotic sensitivity test through Hp culturing in vitro. Results The Hp eradication rate of standard triplet group, berberine group and bismuth group was 55.0%, 77.5% and 95.0%, repectively, the difference was statistically significant(P<0.05). The eradication rate of berberine group was higher than that of standard triplet group, but lower than that of bismuth group(P<0.05). Meanwhile, the minimum antibacterial concentration of berberine was 12.5 mg/ml in vitro. Conclusion Berberine has antibacterial ability against Hp, which combined with standard triple therapy can improve the eradication rate of Hp.
