摘要 目的探究黄芩苷对急性肺损伤大鼠肺组织炎性因子表达的影响。方法将56只雄性SD大鼠随机分为对照组、黄芩苷组与模型组,每组又细分为1 h、4 h与8 h三个亚组,每组8只,采用腹腔注射LPS(10 mg/kg)建立大鼠肺损伤模型。HE染色观察肺组织病理形态学改变,测定并计算各组大鼠肺湿/干重比,免疫组织化学法测定肺组织NF-κB水平,酶联吸附法检测IL-1β、IL-8及TNF-α水平。结果 HE染色结果显示,模型组大鼠肺组织炎性细胞浸润明显,肺泡结构损坏严重;与模型组相比,同一时间点黄芩苷组大鼠炎性细胞浸润减轻,肺泡结构较为完整。与模型组相比,同一时间点黄芩苷组大鼠肺湿/干比重显著降低,肺组织NF-κB蛋白表达的平均光密度值显著降低,血清IL-1β、IL-8及TNF-α的表达水平显著降低(P<0.05)。结论黄芩苷能够通过抑制肺部炎症反应,降低急性肺损伤大鼠的肺组织损伤程度。 Objective To explore the effect of Baicalin on the expression of inflammatory factors in lung tissue of rats with acute lung injury. Methods 56 male SD rats were randomly divided into control group,baicalin group and model group,and each group was subdivided into three subgroups,1 h group,4 h group and 8 h group,8 rats in each group. Acute lung injury was induced by intraperitoneal injection of LPS(10 mg/kg). HE staining was used to observe the pathomorphological changes of lung tissues. The wet/dry weight ratios were measured and calculated. The levels of NF-kappa B in lung tissues were measured by immunohistochemical method. The levels of IL-1 beta,IL-8 and TNF-αlpha were detected by enzyme-linked adsorption method. Results The HE staining showed that there was the inflammatory cells infiltrated significantly in the lung tissue of the model group,and the alveolar structure was damaged seriously. Compared with the model group,the inflammatory cells infiltrated was less and the alveolar structure was more intact in rats of the baicalin group at the same time point. Compared with the model group,the wet/dry rato of lung and the mean optical density of NF-kappa B protein in lung tissue were significantly decreased,and the levels of IL-1 beta,IL-8 and TNF-α in serum were significantly decreased in the baicalin group at the same time point(P<0.05). Conclusion Baicalin could reduce the degree of lung injury in rats with acute lung injury by inhibiting pulmonary inflammation.
