牙周炎患者牙龈组织、龈沟液中HMGB1表达水平及其与外周血炎症因子的关系

Relationship of the expression levels of HMGB1 in gingival tissues and gingival crevicular fluid with inflammatory factors in peripheral blood of patients with periodontitis

目的分析牙周炎患者牙龈组织、龈沟液高迁移率族蛋白1(HMGB1)表达水平与外周血炎症因子的关系。方法回顾性选择2016年2月至2018年5月西宁市口腔医院收治的30例牙周炎作为观察组,选择同期来体检的20例健康志愿者作为对照组。均采集牙龈组织、龈沟液标本,采用免疫组化染色及酶联免疫吸附法(ELISA)测定标本HMGB1表达水平,并留取患者血清标本,采用ELISA法测定血清白介素-1β(IL-1β)、白介素-6(IL-6)、白介素-10(IL-10)、肿瘤坏死因子-α(TNF-α)水平。采用相关性分析法分析牙周炎患者标本HMGB1表达与外周血IL-1、IL-6、IL-10、TNF-α水平的关系。结果观察组牙龈组织HMGB1表达阳性率、龈沟液HMGB1及外周血IL-1β、IL-6、TNF-α表达水平均高于对照组,差异有统计学意义(P <0. 05);牙周炎患者牙龈组织、龈沟液HMGB1表达水平与外周血IL-1β、IL-6、TNF-α水平均呈正相关(P <0. 05)。结论牙周炎患者牙龈组织、龈沟液HMGB1均呈高表达,且与外周血炎症因子IL-1、IL-6、TNF-α表达呈正相关。

Objective To analyze the relationship of the expression levels of high mobility group 1 protein(HMGB1)in gingival tissues and gingival crevicular fluid with inflammatory factors in peripheral blood of patients with periodontitis.Methods 30 patients wit periodontitis admitted to the hospital from February 2016 to May 2018 were selected as the observation group.20 healthy volunteers who underwent physical examination in the same time were used as the control group.Gingival tissue and gingival crevicular fluid specimen were collected.The immumohistochemical staining and enzyme linked immunosorbent assay(ELISA)were performed to determine the expression level of HMGB1 in the specimen.The serum specimens were taken.The levels of serum interleukin-1β(IL-1β),interleukin-6(IL-6),interleukin-10(IL-10)and tumor necrosis factor-α(TNF-α)were determined by ELISA.The correlation analysis was performed to analyze the relationship of expression of HMGB1 in the specimens with the expressions of IL-1,IL-6,IL-10 and TNF-αin peripheral blood of patients with periodontitis.Results The positive rate of HMGB1 expression in gingival tissues,HMGB1 in gingival crevicular fluid and levels of IL-1β,IL-6 and TNF-αin peripheral blood of the observation group were higher than those of the control group(P<0.05).The expression levels of HMGB1 in gingival tissues and gingival crevicular fluid were positively correlated with the levels of IL-1β,IL-6 and TNF-αin peripheral blood(P<0.05).Conclusion The HMGB1 in gingival tissues and gingival crevicular fluid of patients with periodontitis is highly expressed.And it is positively correlated with the expressions of inflammatory factors such as IL-1,IL-6 and TNF-αin peripheral blood.
