槲皮素抗心肌细胞缺氧/复氧损伤的作用及机制研究 被引量:5

摘要 目的研究槲皮素对心肌缺血再灌注损伤的保护作用,初步研究其相关机制。方法构建H9c2心肌细胞缺氧复氧损伤模型,缺氧3 h复氧2 h后,酶标仪检测细胞存活率和乳酸脱氢酶(LDH)、一氧化氮(NO)的含量,Western blotting检测细胞中一氧化氮合酶(NOS)的表达。结果槲皮素能明显降低缺氧复氧后心肌细胞LDH的释放,改善细胞存活率,增加eNOS的表达,减少iNOS的表达,显著降低细胞内NO的含量。结论槲皮素能够影响eNOS和iNOS的表达,进而维持NO生理水平浓度,同时减少细胞损伤甚至凋亡,发挥对心肌缺血再灌注损伤的保护作用。 Objective To investigate the effects of quercetin in cardiomyocytes from anoxia/reoxygenation(A/R)injury,and to further explore the underlying mechanisms involved.Methods The model of H9c2 cell A/R injury was employed to determine whether quercetin protects against myocardial ischemia-reperfusion injury.After reoxygenation for 2 hours,the concentration of NO and the activities of LDH in H9c2 cell were tested by micro plate spectrophotometer.The cell viability was assayed by MTT colorimetric assay.Western blotting analysis measured the expression of NOS.Results The pretreatment of quercetin decreased the release of LDH and increased the cell viability in H9c2 cells undergoing A/R injury.Compared with A/R group,preprocessing 20μM quercetin was significantly decreased expression of iNOS and increased expression of eNOS,and suppressed the produce of NO of H9c2 cells undergoing A/R injury.Conclusions Quercetin plays protection effects during acute A/R injury in H9c2 cell by decreasing the expression of iNOS,increasing expression of eNOS and maintaining physiological concentration of NO.
