马铃薯与枸杞植物组织石蜡切片双重染色技术的改进 被引量:7

摘要 目的找到番红-固绿染色法对不同植物组织石蜡切片染色效果最好的染液配置方法及处理时间,为后续植物组织细胞学研究提供试验基础。方法主要从染液配置方法和酒精脱色时间两个方面来探索出最适于本试验中植物组织番红-固绿的染色法。结果当脱蜡洗去番红浮色及复水透明时间达到了12 min,且番红为乙醇溶液配制时,植物组织未被染色。当脱蜡洗去番红浮色及复水透明时间为6 min左右且番红为乙醇溶液配制时,植物组织只被染为绿色。当脱蜡为6 min,洗去番红,固绿浮色时间较短,且番红为水溶液配制时,植物组织既有红色又有绿色,即染色正常。结论用蒸馏水配制番红染液,且洗去番红浮色固绿染色时间应尽量缩短,避免脱色时间过长,才能获得染色良好的植物组织切片。 Objective To find the best preparation method and treatment time of the staining effect of the red-solid green staining method on the paraffin sections of different plant tissues,and provide the experimental basis for the subsequent plant tissue cytology research. Methods The dyeing method which is most suitable for the plant tissue red-solid green in this experiment was mainly explored from two aspects of the dye preparation method and the treatment time. Results When the time for dewaxing wash-off saffron floating color and rehydration transparent reached 12 min,the sap was prepared in ethanol solution,and the treated plant tissue was not dyed.When the time for dewaxing wash-away the reddish color and the rehydration transparent was about 6 minutes,and the sap was prepared by the ethanol solution,the treated plant tissue was only dyed green.When the dewaxing time is 6 min,the reddish color was washed away,and the solid green coloring time was short.And when the reddish was prepared as an aqueous solution,the treated plant tissue was both red and green,i.e.,the dyeing was normal. Conclusion Preparation of the red dyeing solution with distilled water,and washing of the color of the reddish-colored solid green dye should be as little as possible to avoid washing off the dyed color of the plant tissue,Which helps to obtain the well-stained plant tissue section.
