扶正利湿抗癌方含药血清对前列腺癌PC-3细胞增殖的影响及机制研究 被引量:3

摘要 目的:探讨扶正利湿抗癌方含药血清对前列腺癌细胞株PC-3体外增殖能力的影响及其作用机制。方法:应用CCK-8法测定不同浓度扶正利湿抗癌方含药血清对PC-3细胞生长抑制情况,流式细胞仪检测其对PC-3细胞凋亡及细胞周期的影响,并以Western blot分析扶正利湿抗癌方含药血清对PC-3细胞凋亡调控蛋白bcl-2和bax蛋白表达的影响。结果:扶正利湿抗癌方含药血清对PC-3细胞体外增殖抑制作用明显,呈浓度及时间依赖性。流式结果显示,扶正利湿抗癌方含药血清可诱导PC-3细胞凋亡,并能够增加G_2/M期细胞比例,降低S期细胞比例,呈浓度依赖性。Western blot结果显示,扶正利湿抗癌方含药血清可下调bcl-2蛋白的表达,而上调bax蛋白的表达,呈浓度依赖性。结论:扶正利湿抗癌方含药血清可以通过调节bcl-2和bax蛋白的表达发挥体外抑制PC-3细胞增殖的作用。 Objective: To explore the effects of Fuzheng Lishi Tang on the proliferatio of prostate cancer PC-3 cell lines and exploring the mechanism. Methods: The inhibitory effects of Fuzheng Lishi Tang on growth of PC-3 cells were detected by using CCK-8 assay. The influence of Fuzheng Lishi Tang on apoptosis and cell cycle of PC-3 cells was detected by flow cytometer. The influence of Fuzheng Lishi Tang on protein expressions of bcl-2 and bax of PC-3 cells was analyzed by Western blotting method. Results: The Fuzheng Lishi Tang had inhibitory effect on proliferation of PC-3 cells, its effective time and dose were positively correlated to cellular proliferation inhibition rate. The FCM results demonstrated that the Fuzheng Lishi Tang blocked the cell cycle in the G 2/M phase.Western blot results showed that the Fuzheng Lishi Tang could down-regulate protein expressions of bcl-2 and up-regulate the expression bax of PC-3 cells. Conclusion: The Serum Containing Fuzheng Lishi Tang can inhibit the proliferation of PC-3 cells in vitro by regulating the expression of Bcl-2 and Bax proteins.
