参麻益智方对双侧颈动脉结扎大鼠海马认知功能的影响 被引量:6

The Influence of Shenma Yizhi Decoction on Hippocampal Cognitive Function in Rats with Bilateral Carotid Artery Ligation

摘要 目的观察参麻益智方对双侧颈动脉结扎大鼠行为学及星形胶质细胞和小胶质细胞的影响。方法将60只Wistar大鼠随机分为假手术组(Sham)、双侧颈动脉结扎组(2-VO)、多奈哌齐组(0.5 mg/kg)、中药低剂量组(3.3 mg/kg)、中药中剂量组(6.6 mg/kg)和中药高剂量组(16.5 mg/kg),每组10只。灌胃给药4周后进行水迷宫实验,并观察海马CA1区星形胶质细胞及小胶质细胞的形态改变。结果 2-VO组大鼠逃避潜伏期较Sham组明显延长(P<0.05),目标象限停留时间百分比明显下降(P<0.05);多奈哌齐组及中药高剂量组、中剂量组大鼠逃避潜伏期较2-VO组明显缩短(P<0.05),目标象限停留时间百分比显著延长(P<0.05)。2-VO组大鼠海马CA1区星形胶质细胞及小胶质细胞较Sham组增多,胞体增大。给药后,多奈哌齐组与中药高剂量组、中剂量组星形胶质细胞及小胶质细胞较2-VO组数量减少,胞体变小。结论参麻益智方可改善大鼠学习记忆能力,减轻双侧颈动脉硬化大鼠海马CA1区星形胶质细胞和小胶质细胞的增生和激活,从而改善大鼠认知功能。 Objective To explore the Influence of Shenma Yizhi (SMYZ) decoction on behavioral alteration and the morphological changes of astrocyte and microglia in rats with bilateral carotid artery ligation. Methods sixty male Wistar rats were randomly assigned to Sham,bilateral common carotid arteries occlusion (2-VO),donepezil,SMYZ-Low,Medium and High dose groups with ten rats in each group.Intragastric administration were given respectively for one month then followed Morris water maze test.The morphological alternations of astrocyte and microglia in the CA1 region of hippocampus were observed. Results The 2-VO group showed an elongated escape latency and shortened time spending in target quadrant compared with that in Sham group ( P <0.05).After treatment of donepezil the escape latency significantly reduced in high or medium dose of SMYZ( P <0.05),in coordination with increased time spending in target quadrant ( P <0.05).The amounts of astrocyte and microglia increased remarkably after 2-VO operation as well as enlarged cell body.The donepezil,high and medium dose of SMYZ alleviated these deficits. Conclusion SMYZ decoction can ameliorate the activation and proliferation of astrocyte and microglia,thus improving the cognitive function in 2-VO rats.
