黄连素对APP/PS1转基因小鼠学习记忆能力及淀粉样蛋白表达的影响 被引量:3

摘要 目的探讨黄连素对APP/PS1转基因小鼠学习记忆能力及β-淀粉样蛋白(Aβ)产生和积聚的影响。方法将20只APP/PS1转基因小鼠随机分为生理盐水组和黄连素组,每组10只,适应性喂养后连续灌胃4周。灌胃结束后行水迷宫实验检测两组小鼠学习记忆能力;采用免疫组化及Western blot检测Aβ42蛋白表达;免疫荧光方法检测Aβ积聚形成的老年斑。结果水迷宫定位航行实验结果显示,黄连素组逃避潜伏期较生理盐水组明显缩短(P<0.05);空间探索实验显示,黄连素组穿越原平台位置次数较生理盐水组明显增加(P<0.05),目标象限内活动时间黄连素组长于生理盐水组(P<0.01)。免疫组化及Western blot检测结果显示,黄连素组Aβ42表达水平较生理盐水组降低(P<0.05);免疫荧光结果显示,黄连素组老年斑数量较生理盐水组减少(P<0.05)。结论黄连素能明显改善APP/PS1转基因小鼠学习记忆能力,减少Aβ42的表达和老年斑的形成。 Objective To investigate the influence of berberine on learning and memory ability and β-amyloid production and accumulation in APP/PS1 transgenic mice. Methods Twenty APP/PS1 transgenic mice were randomly divided into normal saline group and berberine group,with 10 mice in each group,and were orally administered for 4 weeks after adaptive feeding.The water maze test was used to observe the learning and memory ability of the two groups.The expression of Aβ42 protein was detected by immunohistochemistry and Western blot.The senile plaque formed by Aβ accumulation was detected by immunofluorescence. Results The water labyrinth navigation experiment showed that the escape latency of the berberine group was significantly shorter than that of the saline group ( P <0.01).The space exploration experiment showed that the number of berberine group crossing the original platform was significantly increased compared with that in the saline group ( P < 0.05).The target quadrant in berberine group was longer than that in the saline group ( P <0.01).Immunohistochemistry and Western blot showed that the expression level of Aβ42 in berberine group was lower than that in normal saline group ( P <0.05).The immunofluorescence showed that the number of senile plaques in berberine group was lower than that in normal saline group ( P <0.05). Conclusion Berberine can effectively improve the learning and memory ability of APP/PS1 transgenic mice,and reduce the expression of Aβ42 and the formation of senile plaques.
