贵州都匀一种聚筛蕊地衣粗多糖的提取及抑菌活性初探 被引量:1

Extraction and antimicrobial activity of crude polysaccharide of Cladia aggregate(Sw.) Ny1 from Duyun in Guizhou

摘要 目的对采自贵州都匀斗篷山景区的一种枝状地衣聚筛蕊进行粗多糖的提取并开展抑菌活性研究,为寻找聚筛蕊地衣天然抗菌成分及该地衣的开发利用提供依据。方法通过形态学对地衣进行鉴定;用水提醇沉法提取地衣的粗提物,用苯酚-硫酸法测定粗提物的含量;通过琼脂平板打孔法评价该粗提物对10种供试菌株的抗菌活性。结果该地衣经形态学特征鉴定为聚筛蕊Cladia aggregate(Sw.) Ny1.;在料液比1∶30(m:v)、温度80℃、次数3次、时间为3 h的条件下,粗多糖含量为19.64 mg/g;抑菌实验结果显示该粗多糖提取物对10种供试菌均不显示抑菌活性。结论首次开展都匀斗篷山景区一种聚筛蕊地衣的粗多糖提取及抗菌活性研究,为该物种的开发利用提供科学依据。 Objective To investigate the extraction and antimicrobial activity of crude polysaccharide of Cladia aggregata(Sw.) Ny1 from Doupeng Mountain Tourist Spots in Duyun of Guizhou and then provide basis for further development.Methods C.aggregate was identified by morphological characteristics under stereomicroscope.Crude polysaccharide was obtained by water extraction and ethanol precipitation method.The content of crude polysaccharide of C.aggregate was determined by phenol-sulfuric acid method.The antimicrobial activity of ten strains was evaluated by age diffusion method.Results C.aggregate was identified according to the morphological characteristics. The content of crude polysaccharide was 19.64 mg/g on the conditions:the ratio of liquid to material being 1∶30(m:v),extraction temperature 80 ℃,frequency of 3 times,and extraction time of 3 h.A antibacterial experiment showed that the crude polysaccharide exhibited no inhibition against the tested ten organisms.Conclusion The extraction and antimicrobial activity of polysaccharides of C.aggregate from Doupeng Mountain Tourist Spots in Duyun of Guizhou were studied for the first time,which might provide scientific basis for further development.
