摘要
目的:通过对地榆药材不同溶剂提取物对肝癌HepG2细胞的抑制作用的比较与作用机制研究,明确地榆药材抗肝癌活性有效组分及其可能的作用机制。方法:在细胞体外培养技术的基础上,使用MTT法对地榆不同溶剂的提取物的活性进行筛选。应用流式细胞仪检测地榆有效组分对细胞凋亡的影响,并采用RT-RCR法检测其对Bax、Bcl-2、caspase-3 mRNA表达的影响。结果:地榆药材的不同溶剂提取物的体外药效具有一定的差异性,其中地榆乙酸乙酯物提取对肝癌细胞的抑制率最大,抑制率为68.30%。流式细胞仪结果显示,地榆乙酸乙酯物提取具有促进细胞凋亡的作用。RT-PCR结果表明,地榆乙酸乙酯提取物能降低肝癌HepG2细胞中Bcl-2 mRNA的表达,增加Bax、caspase-3 mRNA的表达。结论:地榆乙酸乙酯提取物具有体外抗肿瘤作用,其作用机制可能跟其可以促进细胞凋亡有关。该研究明确地榆药材抗肿瘤作用的物质基础,为地榆药材的进一步研究奠定基础。
Objective:Throught comparing inhibitory effect and mechanism of different solvent extract of Sanguisorba offcinalis L. on HepG2 cells,to study its active effective components and potential mechanism. Methods:HepG2 cells were cultured with culture solution,MTT assay was used to examine the proliferation,Annexin V-FITC/PI double staining method was used for the apoptotic rate in HepG2 and RTPCR method to detect the expression of Bax,Bcl-2,caspase-3 mRNA. Results:The inhibitory rate of ethyl acetate extracts from Sanguisorba offcinalis L.(SEAA)on HepG2 cells was 68.30%. SEAA can induce the apoptosis of HepG2 cells,downregulate the expression of Bcl-2 mRNA,and upregulate the expression of Bax,caspase-3 mRNA. Conclusion:SEAA can inhibit the HepG2 cells in vitro by inducing cell apoptosis.In this study,the material basis of the antitumor action of Sanguisorba offcinalis L. was clearly defined,which laid the foundation for further research on Sanguisorba offcinalis L..
