沙参麦冬汤加味对糖尿病大鼠胰岛素抵抗、炎症反应和氧化应激反应的影响 被引量:17

Effect of Modified Shashen Maidong Decoction on Insulin Resistance,Inflammatory Reaction and Oxidative Stress in Diabetic Rats

摘要 【目的】观察沙参麦冬汤加味对糖尿病大鼠胰岛素抵抗、炎症反应和氧化应激反应的影响。【方法】将40只大鼠随机分为空白组、模型组、中药组和西药组,每组10只。除空白组外,其余3组均采用链脲佐菌素腹腔注射结合高脂高糖饮食法建立糖尿病模型。中药组给予沙参麦冬汤加味(剂量为9.4 g·kg-1·d-1)灌胃,西药组给予罗格列酮(4.0 mg·kg-1·d-1)灌胃,空白组和模型组给予等体积的生理盐水灌胃,给药时间为8周。造模结束后,称大鼠体质量,采用全自动生化分析仪检测总胆固醇(TC)、甘油三酯(TG);酶联免疫吸附分析(ELISA)检测血清肿瘤坏死因子α(TNF-α)、白细胞介素(IL)-1β、IL-6、超敏C反应蛋白(hs-CRP)、超氧化物歧化酶(SOD)、丙二醛(MDA)、过氧化氢酶(CAT)、胰岛素(Fins)水平;血糖仪检测空腹血糖(FBG);亲和色谱法检测糖化血红蛋白(HbA1c);胰岛素抵抗指数(HOMA-IR)=FBG×Fins/22.5。【结果】与空白组比较,模型组TC、TG、FBG、Fins、HOMA-IR、HbA1c、TNF-α、IL-1β、IL-6、hs-CRP、MDA水平明显升高,大鼠体质量及SOD、CAT水平明显降低;与模型组比较,中药组TC、TG、FBG、Fins、HOMA-IR、HbA1c、TNF-ɑ、IL-1β、IL-6、hsCRP、MDA水平明显降低,大鼠体质量及SOD、CAT水平明显升高。【结论】沙参麦冬汤加味具有降糖作用,其机制可能与减轻胰岛素抵抗、抑制炎症反应和氧化应激反应有关。 Objective To observe the effects of modified Shashen Maidong Decoction on insulin resistance,inflammatory reaction and oxidative stress in diabetic rats. Methods Forty rats were randomly divided into blankgroup,model group,Chinese medicine group and western medicine group. Except for the control group,the ratsin the other three groups were given intraperitoneal injection of streptozotocin combined with high-fat and high-sugar diet to establish diabetes mellitus model. The Chinese medicine group was given intragastric administration ofmodified Shashen Maidong Decoction at the dosage of 9.4 g · kg^-1· d^-1, western medicine group was givenintragastric administration of rosiglitazone at the dosage of 4 mg·kg^-1·d^-1,the blank group and model group weregiven the same volume of normal saline, the treatment covering 8 weeks. After modeling, the levels of totalcholesterol(TC)and triglyceride(TG)were measured by automatic biochemical analyzer, the levels of tumornecrosis factor α(TNF-α), interleukin(IL)-1β, IL-6, hypersensitive c-reactive protein(hs-CRP),superoxide dismutase(SOD), malondialdehyde(MDA), catalase(CAT), and serum insulin(Fins)weredetected by enzyme-linked immunosorbent assay(ELISA),fasting blood glucose(FBG)level was measured byglucometer, and glycosylated hemoglobin(HbA1 c)level was determined by affinity chromatography. Insulinresistance index(HOMA-IR)=FBG×Fins/22.5. Results Compared with the blank group,the levels of TC,TG,FBG, Fins, HOMA-IR, HbA1 c, TNF-α,IL-1β, IL-6, hs-CRP and MDA in the model group weresignificantly increased, and the rat body mass, the levels of SOD and CAT were significantly decreased.Compared with the model group,the levels of TC,TG,FBG,Fins,HOMA-IR,HbA1 c,TNF-α,IL-1β,IL-6,hs-CRP and MDA were significantly decreased in Chinese medicine group,and the rat body mass,thelevels of SOD and CAT were significantly increased. Conclusion Modified Shashen Maidong Decoction has certaineffect on lowering glucose, and its mechanism may be related with alleviating insulin resistance, inhibitinginflammatory reaction and oxidative stress.
